大蒜油聯(lián)合5-FU誘導(dǎo)腺樣囊性癌細(xì)胞株ACC-M凋亡的研究

【摘要】:目的:探討大蒜油聯(lián)合5-FU誘導(dǎo)腺樣囊性癌細(xì)胞株ACC-M凋亡的效應(yīng)。方法:人涎腺腺樣囊性癌細(xì)胞株ACC-M細(xì)胞培養(yǎng),分為實(shí)驗(yàn)組(5-FU組、大蒜油組、大蒜油+5-FU組)和對照組,采用MTT法觀察腫瘤細(xì)胞的體外生長活性;采用流式細(xì)胞術(shù)分析腫瘤細(xì)胞周期和凋亡率的變化。結(jié)果:MTT實(shí)驗(yàn)結(jié)果顯示不同濃度的5-FU、大蒜油、大蒜油+5-FU對ACC-M細(xì)胞均有抑制作用,隨著濃度和作用時(shí)間的增加而呈上升趨勢;流式細(xì)胞術(shù)分析顯示細(xì)胞周期發(fā)生明顯變化,隨著濃度增加、作用時(shí)間延長G0/G1期的細(xì)胞比率上升,S期無明顯變化,而G2/M期的細(xì)胞下降,凋亡率顯示大蒜油聯(lián)合5-FU誘導(dǎo)ACC-M細(xì)胞凋亡明顯強(qiáng)于單獨(dú)組。結(jié)論:大蒜油能有效誘導(dǎo)腺樣囊性癌細(xì)胞株ACC-M凋亡,大蒜油聯(lián)合5-FU應(yīng)用對ACC-M細(xì)胞的作用明顯強(qiáng)于大蒜油,5-FU可單獨(dú)應(yīng)用。
[Abstract]:Objective: to investigate the effect of garlic oil combined with 5-FU on apoptosis of adenoid cystic carcinoma cell line ACC-M. Methods: human salivary adenoid cystic carcinoma cell line ACC-M was cultured and divided into experimental group (5-FU group, garlic oil 5-FU group) and control group. The growth activity of tumor cells in vitro was observed by MTT assay. The changes of tumor cell cycle and apoptosis rate were analyzed by flow cytometry. Results: the results of MTT assay showed that garlic oil 5-FU could inhibit ACC-M cells with different concentrations of 5 Fu and garlic oil FU, and increased with the increase of concentration and time. Flow cytometry analysis showed that the cell cycle changed obviously. With the increase of concentration, the cell ratio in G0/G1 phase increased, but there was no significant change in S phase, but decreased in G2 鈮,
