牽張應(yīng)力對(duì)人牙周膜細(xì)胞NF-κB信號(hào)通路的影響及其與MMP-2表達(dá)的關(guān)系

【摘要】:目的:通過(guò)膠原酶消化法體外分離培養(yǎng)人牙周膜細(xì)胞并構(gòu)建細(xì)胞牽張應(yīng)力加載模型,研究牽張應(yīng)力對(duì)人牙周膜細(xì)胞NF-κB信號(hào)通路的影響及其與MMP-2表達(dá)的關(guān)系. 方法:①膠原酶消化法體外分離培養(yǎng)人牙周膜細(xì)胞,觀察人牙周膜細(xì)胞生物學(xué)特性,免疫組織化學(xué)對(duì)細(xì)胞進(jìn)行抗角蛋白及抗波形蛋白抗體檢測(cè)判定其來(lái)源,并構(gòu)建細(xì)胞牽張應(yīng)力加載模型。 ②選取4-6代細(xì)胞,對(duì)細(xì)胞施加不同時(shí)間的12%周期性牽張應(yīng)力,結(jié)合信號(hào)通路抑制劑及蛋白印跡的檢測(cè)方法,研究不同應(yīng)力條件對(duì)人牙周膜細(xì)胞NF-κB、IκB、 MMP-2蛋白的表達(dá)、IKK激酶的活性及磷酸化水平的影響,及MMP-2在人牙周膜細(xì)胞中表達(dá)的可能的信號(hào)轉(zhuǎn)導(dǎo)通路。 結(jié)果:①膠原酶消化法體外成功分離培養(yǎng)人牙周膜細(xì)胞,免疫組織化學(xué)鑒定其來(lái)源可靠;應(yīng)用多通道細(xì)胞牽張應(yīng)力加載系統(tǒng)成功構(gòu)建人牙周膜細(xì)胞體外培養(yǎng)-力學(xué)刺激模型。 ②蛋白印跡結(jié)果顯示:人牙周膜細(xì)胞受到12%形變率的周期性牽張應(yīng)力作用時(shí),,NF-κB信號(hào)通路被激活, p-IKK磷酸化程度升高,與對(duì)照組相比均具有顯著性差異(P0.05)。PDTC組, NF-κB蛋白、p-IKK表達(dá)明顯降低,與12h組相比具有顯著性差異(P0.05)。對(duì)照組胞漿I-κBα蛋白有較強(qiáng)的表達(dá),加力后,胞漿I-κBα蛋白發(fā)生磷酸化并降解表達(dá)顯著降低與對(duì)照組相比具有顯著差異(P0.05),加入PDTC后,胞漿I-κBα蛋白表達(dá)增加,與12h相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 ③蛋白印跡結(jié)果顯示:當(dāng)牙周膜細(xì)胞受到12%形變率的周期性牽張應(yīng)力作用時(shí),MMP-2蛋白的表達(dá)增加,與對(duì)照組相比具有顯著性差異(P0.05),12h時(shí)MMP-2蛋白表達(dá)最高。NF-κB通路抑制劑PDTC可抑制機(jī)械牽張應(yīng)力作用下牙周膜細(xì)胞MMP-2表達(dá)的增加,與單純加力組相比差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論:①采用膠原酶消化法成功的分離培養(yǎng)人牙周膜細(xì)胞并構(gòu)建hPDLCs-力學(xué)刺激模型。 ②12%形變率的周期性牽張應(yīng)力作用于牙周膜細(xì)胞時(shí),NF-κB通路被激活,胞漿I-κBα蛋白表達(dá)水平的降低,I-κBα降解是NF-κB信號(hào)通路激活的機(jī)制之一。 ③12%形變率的周期性機(jī)械牽張應(yīng)力可誘導(dǎo)人牙周膜細(xì)胞MMP-2的表達(dá)增加,從而影響牙周組織細(xì)胞外基質(zhì)代謝同時(shí)機(jī)械牽張應(yīng)力可通過(guò)NF-κB通路影響MMP-2蛋白的表達(dá)。
