PTH和SDF-1聯(lián)合應(yīng)用對(duì)人牙周膜干細(xì)胞增殖、遷移及分化的影響

【摘要】:目的:基質(zhì)細(xì)胞衍生因子-1(stromal cell derived factor-1,SDF-1)是原位組織工程中最重要的趨化因子之一,對(duì)人牙周膜干細(xì)胞有趨化和促增殖分化作用,局部應(yīng)用能夠促進(jìn)牙周組織再生。甲狀旁腺激素(parathyroid hormone, PTH)作為唯一得到美國(guó)食品和藥物管理局(Food and Drug Administration, FDA)批準(zhǔn)的用于治療骨質(zhì)疏松的骨合成代謝藥物,能夠動(dòng)員骨髓基質(zhì)細(xì)胞進(jìn)入血液,并抑制蛋白二肽基肽酶-Ⅳ(dipeptidyl peptidase-Ⅳ, DPP-Ⅳ)對(duì) SDF-1的降解。目前PTH和SDF-1聯(lián)合應(yīng)用促進(jìn)組織再生的研究更多的是集中心臟損傷方面,應(yīng)用于牙周組織再生方面的研究甚少。本研究的目的是探討PTH和SDF-1聯(lián)合應(yīng)用對(duì)人牙周膜干細(xì)胞增殖、遷移及分化的影響,為牙周組織再生的深入研究提供實(shí)驗(yàn)依據(jù)。方法:從因正畸需要而拔除的健康前磨牙上分離、培養(yǎng)人牙周膜干細(xì)胞(human periodontal ligament stem cells, hPDLSCs),用有限稀釋法篩選單細(xì)胞克隆;通過(guò)細(xì)胞活力檢測(cè)試劑盒(cell-counting kit-8, CCK8)、細(xì)胞遷移試驗(yàn)、堿性磷酸酶(Alkaline phosphatase, ALP)活性測(cè)試、茜素紅染色法和實(shí)時(shí)定量聚合酶鏈反應(yīng)(quantitative real-time polymerase chain reaction, qRT-PCR)測(cè)定PTH與SDF-1聯(lián)合應(yīng)用對(duì)hPDLSCs增殖、遷移能力和成骨分化能力的影響。使用SPSS 16.0統(tǒng)計(jì)軟件進(jìn)行單因素方差分析,數(shù)據(jù)均用Mean±SEM表示。P0.05具有統(tǒng)計(jì)學(xué)差異。結(jié)果:①成功分離培養(yǎng)出人牙周膜干細(xì)胞,細(xì)胞形態(tài)正常,狀態(tài)良好。②PTH與SDF-1聯(lián)合應(yīng)用能夠促進(jìn)PDLSCs的增殖和遷移能力:CCK8結(jié)果表明PTH與SDF-1聯(lián)合應(yīng)用促進(jìn)細(xì)胞增殖的效應(yīng)最強(qiáng)(P0.05);遷移實(shí)驗(yàn)表明實(shí)驗(yàn)組均能夠促進(jìn)牙周膜干細(xì)胞的遷移(P0.05),聯(lián)合PTH之后使SDF-1的促遷移效應(yīng)更強(qiáng)(P0.05)。③PTH與SDF-1聯(lián)合應(yīng)用能夠促進(jìn)PDLSCs的成骨分化:ALP活性測(cè)試結(jié)果是細(xì)胞培養(yǎng)7天時(shí)PTH與SDF-1聯(lián)合應(yīng)用能夠顯著增加ALP活性(P0.05),細(xì)胞培養(yǎng)14天時(shí)實(shí)驗(yàn)組與對(duì)照組之間的ALP活性已無(wú)顯著性差異(P0.05),表明PTH與SDF-1聯(lián)合應(yīng)用在細(xì)胞培養(yǎng)的早期能增加ALP活性;茜素紅染色結(jié)果表明PTH與SDF-1聯(lián)合應(yīng)用能夠顯著促進(jìn)礦化結(jié)節(jié)的形成CP0.05);qRT-PCR結(jié)果顯示PTH與SDF-1聯(lián)合應(yīng)用能夠上調(diào)成骨相關(guān)標(biāo)志物BSP、Runx2、OCN的基因表達(dá)水平(P0.05)。結(jié)論:本實(shí)驗(yàn)證實(shí)PTH與SDF-1聯(lián)合應(yīng)用能夠更大程度的促進(jìn)人牙周膜干細(xì)胞增殖、遷移及成骨分化。
