A-NK細胞抗口腔舌鱗狀細胞癌的實驗研究

【摘要】:目的觀察黏附性自然殺傷(A-NK)細胞體外生長與增殖能力,探討其治療裸鼠舌鱗狀細胞癌移植瘤的效果及其作用機制,為臨床治療鱗癌提供理論依據(jù)。方法抽取健康人外周血,分選出A-NK細胞和非黏附性自然殺傷(NA-NK)細胞體外培養(yǎng),鏡下觀察細胞生長情況。建立裸鼠舌鱗癌皮下移植瘤模型,隨機分成3組,每組7只小鼠,分別瘤旁注射生理鹽水、A-NK細胞和NA-NK細胞,觀察腫瘤體積變化,33 d后,處死裸鼠,剝離移植瘤,稱瘤質(zhì)量,繪制腫瘤生長曲線。結(jié)果鏡下觀察,A-NK細胞在第15天達到增殖高峰,NA-NK細胞于第12天達到增殖高峰。培養(yǎng)3周后,A-NK細胞共增加39.33倍,NA-NK細胞僅增加16.33倍。生理鹽水組裸鼠口腔舌鱗癌移植瘤體積大于NA-NK細胞組和A-NK細胞組,NA-NK細胞組大于A-NK細胞組,3組裸鼠腫瘤體積在組間、時間和交互效應(yīng)上差異均有統(tǒng)計學意義(P0.01)。第33天,生理鹽水組裸鼠瘤質(zhì)量大于NA-NK細胞組和A-NK細胞組,NA-NK細胞組大于A-NK細胞組(P0.05)。結(jié)論 A-NK細胞和NA-NK細胞均可在裸鼠體內(nèi)誘導(dǎo)明顯的抗腫瘤效應(yīng),且A-NK的抑瘤作用大于NA-NK細胞。
[Abstract]:Aim to observe the growth and proliferation of adhesive natural killer (A-NK) cells in vitro, and to explore the therapeutic effect and mechanism of adhensive natural killer cells in nude mice with tongue squamous cell carcinoma (LSCC), so as to provide a theoretical basis for clinical treatment of squamous cell carcinoma (SCC). Methods A-NK cells and non-adhesive natural killer (NA-NK) cells were selected from the peripheral blood of healthy people and cultured in vitro. The growth of the cells was observed under microscope. The model of subcutaneous transplantation of tongue squamous cell carcinoma in nude mice was established. Seven mice in each group were randomly divided into 3 groups. Normal saline, A-NK cells and NA-NK cells were injected into the tumor side to observe the change of tumor volume. 33 days later, nude mice were killed and transplanted tumor was exfoliated. Weigh the mass of the tumor and draw the growth curve of the tumor. Results under microscope, A-NK cells reached the peak of proliferation on the 15th day and NA-NK cells reached the proliferation peak on the 12th day. After 3 weeks of culture, A-NK cells increased 39.33-fold and NA-NK cells only 16.33-fold. The transplanted tumor volume of oral tongue squamous cell carcinoma in saline group was larger than that of NA-NK cell group and A-NK cell group, and that of NA-NK cell group was larger than that of A-NK cell group, and the tumor volume of three groups was between groups. There were significant differences in time and interaction (P0.01). On the 33rd day, the tumor mass of nude mice in saline group was larger than that in NA-NK cell group and A-NK cell group, and that in NA-NK cell group was higher than that in A-NK cell group (P0.05). Conclusion both A-NK cells and NA-NK cells can induce obvious antitumor effect in nude mice, and the antitumor effect of A-NK is greater than that of NA-NK cells.
