應(yīng)用微種植支抗大鼠牙齒移動(dòng)過程中牙周組織OPN的表達(dá)變化

【摘要】:目的觀察應(yīng)用微型種植支抗使大鼠牙齒移動(dòng)過程中骨橋蛋白(OPN)的表達(dá)變化,探討其在成骨細(xì)胞誘導(dǎo)和骨改建中的作用。方法建立應(yīng)用微型種植支抗?fàn)恳苿?dòng)大鼠下頜第一磨牙的實(shí)驗(yàn)?zāi)P汀TM組在大鼠右側(cè)下頜做加力牽引,左側(cè)下頜作為對照組,分別于OTM后2、4、7及14天,解剖分離大鼠下頜骨,HE染色觀察組織形態(tài),免疫組織化學(xué)檢測OPN表達(dá)變化及時(shí)間分布特點(diǎn)。結(jié)果 HE顯示OTM組中張力側(cè)成骨細(xì)胞、壓力側(cè)破骨細(xì)胞表達(dá)較多。免疫組化顯示OTM組OPN陽性細(xì)胞數(shù)隨加力時(shí)間的增加而增加,第7天達(dá)到高峰,此后逐漸降低。結(jié)論在正畸牙移動(dòng)過程中,OPN表達(dá)的變化規(guī)律與骨改建過程中成骨細(xì)胞分化、形成和功能密切相關(guān),與牙槽骨改建過程一致。
[Abstract]:Objective to observe the expression of osteopontin (OPN) in the process of tooth movement in rats induced by mini-implant Anchorage, and to explore the role of osteopontin (OPN) in osteoblast induction and bone remodeling. Methods the experimental model of mandibular first molar movement in rats was established with mini-implant Anchorage. The mandible was dissected from the right mandible in the OTM group and the left mandible was used as the control group. The mandible was dissected on the 7th and 14th days after OTM. Tissue morphology was observed by HE staining and OPN expression and time distribution were detected by immunohistochemistry. Results HE showed that the osteoblasts in the tension side and the osteoclasts in the pressure side were more expressed in the OTM group. Immunohistochemistry showed that the number of OPN positive cells in OTM group increased with the increase of adding time, reached the peak on the 7th day, and then decreased gradually. Conclusion during orthodontic tooth movement, the expression of OPN is closely related to the differentiation, formation and function of osteoblasts during bone remodeling, which is consistent with the process of alveolar bone remodeling.
