力與炎癥因子對牙周膜細胞破骨調(diào)控因子表達的影響

【摘要】:目的觀察力信號和炎癥因子在牙周膜細胞向破骨細胞分化過程中對功能蛋白表達的影響。方法收集因正畸治療需要拔除的無齲前磨牙,體外培養(yǎng)人牙周膜細胞,分別對其施加5%基底型變量的周期性牽張力和白細胞介素(IL)-1β、-6、-23和腫瘤壞死因子(TNF)α等炎癥因子刺激,Western blot檢測施加刺激0、2、4、8、12、24 h后骨代謝相關(guān)調(diào)控因子骨保護素(OPG)和細胞因子κB受體激動劑配體(RANKL)的蛋白表達情況。結(jié)果炎癥因子對牙周膜細胞破骨向因子表達有促進作用,而周期性牽張力能夠抑制破骨調(diào)控因子的表達,兩者共同作用時破骨向因子呈現(xiàn)高表達狀態(tài)。結(jié)論炎癥因子對破骨向調(diào)控因子有促進作用,而周期性牽張力抑制破骨的作用無法抵消這一過程。
[Abstract]:The effects of observation signals and inflammatory factors on the expression of functional proteins during the differentiation of periodontal ligament cells into osteoclasts. Methods Human periodontal ligament cells were cultured in vitro and subjected to cyclic tension of 5% basal variables and interleukin (IL) 1 尾, -6, respectively, which were extracted for orthodontic treatment. The expression of osteoprotegerin (OPG) and cytokine 魏 B receptor agonist ligand (RANKL) (RANKL) and osteoprotegerin (OPG), a regulatory factor related to bone metabolism, were detected by, Western blot stimulated by -23 and tumor necrosis factor (TNF) 偽 at 1224 h after stimulation. Results inflammatory factors could promote the expression of osteoclasts in periodontal ligament cells, and cyclic tension could inhibit the expression of osteoclast regulatory factors. Conclusion inflammatory factors can promote osteoclast regulation factor, but cyclic tension can not counteract this process.
