納米銀對變異鏈球菌標(biāo)準(zhǔn)株及耐氟株抗菌性的研究

【摘要】:目的對比葡萄糖和次磷酸鈉分別還原硝酸銀制備兩種納米銀的性能,檢測兩種納米銀對變異鏈球菌標(biāo)準(zhǔn)株及耐氟株的抑菌圈直徑、最小抑菌濃度(MIC)、最小殺菌濃度(MBC),在兩種納米銀對實(shí)驗(yàn)菌的抗菌效果方面進(jìn)行初步研究。方法采用葡萄糖和次磷酸鈉分別還原硝酸銀制備兩種納米銀,通過UV-Vis、XRD及TEM三種方法對兩種納米銀進(jìn)行表征;利用變異鏈球菌標(biāo)準(zhǔn)株以及前期實(shí)驗(yàn)室成功構(gòu)建并保存的變異鏈球菌耐氟株為實(shí)驗(yàn)菌株,制備兩實(shí)驗(yàn)菌株的TPY平板,分別放上兩種納米銀溶膠濾紙片,將平板倒置、放于37℃恒溫箱微需氧過夜培養(yǎng)16h(以下培養(yǎng)條件相同),觀察并測量抑菌圈直徑;取兩塊96孔板,在每一板內(nèi)分別向第一行的1到10孔內(nèi)依次加入100μL二倍稀釋后的銀溶膠,然后吸取100μL稀釋為106CFU/mL的兩菌液分別加到兩板的以上各孔內(nèi),密封孔板,觀察各孔的渾濁度(同理檢測另一種納米銀);從未渾濁的實(shí)驗(yàn)孔里分別取100μL液體,滴到TPY固體培養(yǎng)基上,涂布均勻,觀察并計數(shù)菌板上實(shí)驗(yàn)菌生長情況。測定兩種納米銀對實(shí)驗(yàn)菌株的抑菌圈直徑、最小抑菌濃度(MIC)、最小殺菌濃度(MBC)。 結(jié)果葡萄糖和次磷酸鈉分別制備的納米銀均為:分散性好、穩(wěn)定性較好、均粒徑分別約為13.40nm、15.50nm的球形粒子,且前者的性能優(yōu)于后者;葡萄糖制備的納米銀對標(biāo)準(zhǔn)株及耐氟株的抑菌圈直徑分別為(20.87±0.66) mm、(19.68±0.86)mm,MIC分別為(2.08±0.90)μg/mL、(2.60±0.90)μg/mL, MBC分別為(4.16±1.81)μg/mL、(5.21±1.81)μg/mL、;次磷酸鈉制備的納米銀對標(biāo)準(zhǔn)株及耐氟株的抑菌圈直徑分別為(14.86±O.43)mm.(14.19±0.50)mm, MIC為(35.15±12.18)μg/mL、(56.24±24.36)μg/mL,MBC為(70.30±24.36) μg/mL、(1l2.48±48.72)μg/mL。 結(jié)論葡萄糖和次磷酸鈉分別還原硝酸銀制備了分散性好、穩(wěn)定性較好、均粒徑較小約為13.40nm、15.50nm球形粒子的納米銀;兩種納米銀對變異鏈球菌標(biāo)準(zhǔn)株及耐氟株均有較高的抗菌性,且前者的抗菌性較后者好,但同種納米銀對標(biāo)準(zhǔn)株和耐氟株抗菌性的差異不顯著。
[Abstract]:Objective to compare the properties of glucose and sodium hypophosphate in reducing silver nitrate to prepare two kinds of silver nanoparticles, and to detect the diameter of bacteriostasis circle, minimum inhibitory concentration (MIC),) and minimal bactericidal concentration (MBC),) of two kinds of silver nanoparticles to Streptococcus mutans standard strain and fluorine-resistant strain. The antimicrobial effects of two kinds of silver nanoparticles against experimental bacteria were preliminarily studied. Methods two kinds of silver nanoparticles were prepared by reducing silver nitrate by glucose and sodium hypophosphite respectively. The two kinds of silver nanoparticles were characterized by UV-Vis,XRD and TEM. Using the standard strain of Streptococcus mutans and the fluorine-resistant strain of Streptococcus mutans successfully constructed and preserved in the laboratory, the TPY plates of the two strains were prepared.
