MALDI-TOF質(zhì)譜技術(shù)鑒定齲病患者唾液中的變異鏈球菌

【摘要】:目的:利用基質(zhì)輔助激光解吸電離時(shí)間飛行質(zhì)譜(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF MS)鑒定齲病患者唾液中的變異鏈球菌,為探尋快速、準(zhǔn)確鑒定變異鏈球菌的方法提供依據(jù)。方法:研究對(duì)象為北京大學(xué)口腔醫(yī)院牙體牙髓科就診的90名齲病患者,收集受試者的唾液樣本,處理后進(jìn)行MALDI-TOF MS檢測(cè),結(jié)果導(dǎo)入BioExplorer 1.0軟件與標(biāo)準(zhǔn)庫(kù)比對(duì),分值≥25者鑒定為變異鏈球菌,25者鑒定為非變異鏈球菌。最后將質(zhì)譜和16S rDNA測(cè)序結(jié)果進(jìn)行比對(duì),計(jì)算靈敏度和一致率。結(jié)果:MALDI-TOF MS鑒定變異鏈球菌的靈敏度為96.0%,與16S rDNA測(cè)序結(jié)果的一致率為98.7%。結(jié)論:MALDI-TOF MS是一種高通量、快速且操作簡(jiǎn)便的方法,鑒定變異鏈球菌的靈敏度和一致率都很高,具有較高的臨床應(yīng)用價(jià)值。
[Abstract]:Objective: to identify Streptococcus mutans in saliva of caries patients by matrix assisted laser desorption ionization time flight mass spectrometry (matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF MS). The method of identifying Streptococcus mutans accurately provides the basis. Methods: 90 dental caries patients in dental pulp department of Peking University Stomatology Hospital were studied.
