【摘要】：目的研究牽張應(yīng)變對(duì)人牙周膜細(xì)胞(human periodontal ligament cells,HPDLCs)表達(dá)炎癥體通路相關(guān)因子的影響。方法對(duì)體外培養(yǎng)的HPDLCs施加20%牽張應(yīng)變6、24 h,分別采用實(shí)時(shí)定量PCR以及免疫印跡技術(shù)檢測(cè)NLRP3、Caspase-1及IL-1β的mRNA及蛋白表達(dá)的變化。對(duì)體外培養(yǎng)的HPDLCs施加20%牽張應(yīng)變1、2、4、6 h后,采用酶聯(lián)免疫吸附實(shí)驗(yàn)檢測(cè)細(xì)胞分泌的IL-1β蛋白變化。對(duì)照組HPDLCs培養(yǎng)于彈性培養(yǎng)皿中,與動(dòng)態(tài)牽張應(yīng)變加載組的培養(yǎng)條件一致,但不加載應(yīng)變。結(jié)果 6 h牽張應(yīng)變作用下HPDLCs中NLRP3、caspase-1及IL-1β的mRNA及蛋白表達(dá)較對(duì)照組增加(P0.05);24 h牽張應(yīng)變作用下HPDLCs中NLRP3的蛋白表達(dá)水平上調(diào)(P0.05);加載牽張應(yīng)變4、6 h后,細(xì)胞培養(yǎng)上清中IL-1β含量隨牽張時(shí)間的增長(zhǎng)而增加(P0.05)。結(jié)論20%機(jī)械牽張6 h能誘導(dǎo)HPDLCs中NLRP3/Caspase-1炎癥體-IL-1β通路因子的表達(dá)。
[Abstract]:Objective to study the effect of stretch strain on the expression of inflammatory pathway related factors in human periodontal ligament cells (human periodontal ligament cells,HPDLCs). Methods the expression of mRNA and protein of NLRP3,Caspase-1 and IL-1 尾 were detected by real-time quantitative PCR and Western blotting. The changes of IL-1 尾 protein secreted by HPDLCs were detected by enzyme-linked immunosorbent assay (Elisa) for 6 h after the strain of 20% stretch strain was applied to the cells. The HPDLCs of control group was cultured in elastic petri dish, which was consistent with the culture condition of dynamic strain loading group, but no strain was applied. Results the expression of mRNA and protein of NLRP3,caspase-1 and IL-1 尾 in HPDLCs at 6 h was higher than that in control group (P0.05), and the expression of NLRP3 protein in HPDLCs was up-regulated under 24 h stretch strain (P0.05). The content of IL-1 尾 in the supernatant of cell culture increased with the increase of stretch time (P0.05). Conclusion 20% mechanical stretch for 6 h can induce the expression of IL-1 尾 pathway factor in NLRP3/Caspase-1 inflammatory body in HPDLCs.
