【摘要】：目的:探討舌鱗癌細胞(human tongue squamous cell carcinoma cells,SCC)經(jīng)順鉑化療后的增殖和遷移能力的改變,及其對細胞增殖相關(guān)Nanog基因表達的調(diào)控作用。方法:本文應(yīng)用5、4、3.5μg/m L順鉑分別作用于舌鱗癌細胞系SCC-15,處理時間分別為24、48、72 h(實驗組),未經(jīng)順鉑處理的細胞為對照。流式細胞儀檢測細胞凋亡率,q RT-PCR及Western blot檢測2組細胞中Nanog的表達情況,劃痕實驗檢測2組細胞遷移能力的變化。結(jié)果:經(jīng)順鉑處理后細胞凋亡率增加,48 h后凋亡率顯著增加(P=0.000)。q RT-PCR結(jié)果顯示24、48、72 h組的Nanog基因相對表達量在順鉑處理的細胞中逐漸增加,48 h Nanog基因表達量與對照組相比,差異具有統(tǒng)計學意義(P=0.000)。Western blot實驗結(jié)果進一步驗證了順鉑處理的舌鱗癌細胞中Nanog蛋白相對表達量較對照組高(P≤0.001)。劃痕實驗顯示經(jīng)順鉑作用時間越長的實驗組,細胞遷移速率越高。結(jié)論:順鉑化療對舌鱗癌細胞增殖能力和遷移能力有調(diào)控作用。
[Abstract]:Aim: to investigate the effects of cisplatin chemotherapy on proliferation and migration of tongue squamous cell carcinoma (human tongue squamous cell carcinoma cells,SCC) cells and its regulatory effect on the expression of Nanog gene.
