【摘要】：目的:觀察微溝槽形貌對人牙齦成纖維細(xì)胞(HGFs)黏附形態(tài)和增殖的影響。方法:在鈦表面設(shè)計(jì)不同溝槽尺寸,利用光刻技術(shù)制作微溝槽形貌,溝槽寬度與間隔分別為15、30、60μm,溝槽的深度為5μm和10μm,分組為T15/5,T15/10,T30/5,T30/10,T60/5,T60/10,光滑鈦(T0)表面為對照組,環(huán)境掃描電鏡(ESEM)觀察各組材料表面微溝槽形貌,接種HGFs后,通過掃描電鏡和CCK-8實(shí)驗(yàn)觀察分析不同溝槽尺寸上方細(xì)胞黏附和增殖差異。結(jié)果:T0組表面細(xì)胞排列不規(guī)則,其余組溝槽表面細(xì)胞均順著溝槽排列,T60/10組溝槽表面細(xì)胞的增殖幅度最大,增殖時(shí)間最長。結(jié)論:本實(shí)驗(yàn)所設(shè)計(jì)的溝槽尺寸均能成功誘導(dǎo)細(xì)胞順著溝槽排列,隨著溝槽的寬度增加深度增大越有利于促進(jìn)細(xì)胞增殖。
[Abstract]:Aim: to observe the effect of microgroove morphology on (HGFs) adhesion and proliferation of human gingival fibroblasts. Methods: different grooves were designed on the surface of titanium. The microgrooves were fabricated by photolithography. The width and spacing of the grooves were 1530U / 60 渭 m and 10 渭 m, respectively. The grooves were divided into T 15 / 5 T 15 / 10 T 305 / T 30 / 10 T 605T 60 / 10 and smooth titanium (T 0) surfaces were used as the control group. The morphology of microgrooves was observed by environmental scanning electron microscope (ESEM). After inoculated with HGFs, the adhesion and proliferation of cells above different grooves were observed and analyzed by SEM and CCK-8 experiments. Results the surface cells of the T0 group were arranged irregularly, and the cells of the other groups were all arranged along the grooves. The proliferative amplitude and the time of the cells on the grooves surface of the 10 groups were the largest and the longest. Conclusion: the size of the grooves designed in this experiment can successfully induce the cells to arrange along the grooves, and the increase of the depth with the width of the grooves is beneficial to the proliferation of the cells.
