【摘要】：目的:探討尼古丁對(duì)體外培養(yǎng)人牙齦成纖維細(xì)胞(HGFs)的增殖及其分泌IL-8的影響。方法:組織塊法原代培養(yǎng)HGFs,MTT法觀察10、50、100、250、500、1000、2 000μg/mL不同濃度尼古丁對(duì)其增殖的影響;取第4代HGFs分別與10、50、100、250、500、1000μg/mL不同濃度尼古丁共同培養(yǎng)72 h,用酶聯(lián)免疫吸附(ELISA)法檢測(cè)HGFs培養(yǎng)上清中IL-8的含量。結(jié)果:50~2 000μg/mL不同濃度的尼古丁均能明顯抑制HGFs的增殖(P0.05),且呈濃度依賴性;10~500μg/mL尼古丁均能明顯促進(jìn)HGFs合成IL-8(P0.05),其中以100μg/mL濃度組的促進(jìn)作用最強(qiáng)。結(jié)論:一定濃度尼古丁可抑制HGFs的增殖并促進(jìn)其合成IL-8,提示尼古丁可能通過(guò)調(diào)控IL-8的表達(dá)而造成對(duì)牙周組織的破壞。
[Abstract]:Aim: to investigate the effect of nicotine on (HGFs) proliferation and IL-8 secretion in cultured human gingival fibroblasts. Methods: the effects of different concentrations of nicotine at different concentrations on the proliferation of HGFs were observed by tissue block method, and the content of IL-8 in the supernatant of HGFs culture was detected by enzyme-linked immunosorbent (ELISA) (Elisa) for 72 hours after the fourth generation of HGFs was co-cultured with 1050,500,500,500 渭 g/mL nicotine for 72 hours. The results were as follows: (1) the content of nicotine in the supernatant of HGFs culture was determined by enzyme-linked immunosorbent (ELISA) (Elisa). Results nicotine at different concentrations of 渭 g/mL significantly inhibited the proliferation of HGFs (P0.05), and in a concentration-dependent manner, nicotine (10500 渭 g/mL) significantly promoted the synthesis of IL-8 by HGFs (P0.05), especially in the 100 渭 g/mL group. Conclusion: nicotine can inhibit the proliferation of HGFs and promote its synthesis of IL-8, suggesting that nicotine may damage periodontal tissue by regulating the expression of IL-8.
