【摘要】：目的探討轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)對(duì)癌相關(guān)成纖維細(xì)胞(CAFs)中成纖維細(xì)胞活化蛋白(FAP)表達(dá)的影響。方法應(yīng)用10 ng/ml的TGF-β1作用于CAFs細(xì)胞,設(shè)置正常成纖維細(xì)胞(NFs)和未經(jīng)處理的CAFs細(xì)胞作為對(duì)照組,采用RT-PCR和Western blot法檢測(cè)細(xì)胞中FAP的蛋白和mRNA表達(dá)情況。結(jié)果 FAP在NFs中幾乎不表達(dá),而在CAFs中FAP的mRNA和蛋白表達(dá)均增加。與NFs組和CAFs組相比,10 ng/ml的TGF-β1能夠明顯促進(jìn)CAFs細(xì)胞分泌FAP,作用12、24 h后FAP mRNA和蛋白的相對(duì)表達(dá)量差異有統(tǒng)計(jì)學(xué)意義(P0.05)。且隨著作用時(shí)間的延長(zhǎng),FAP的表達(dá)持續(xù)升高,24 h較12 h表達(dá)增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 TGF-β1能夠促進(jìn)CAFs中FAP的表達(dá),在腫瘤上皮-間質(zhì)交互作用中扮演著重要角色。
[Abstract]:Objective to investigate the effect of transforming growth factor - beta 1 (TGF- beta 1) on the expression of fibroblast activation protein (FAP) in cancer related fibroblasts (CAFs). Methods the TGF- beta 1 of 10 ng/ml was used in CAFs cells, normal fibroblasts (NFs) and untreated CAFs cells were set as the control group. RT-PCR and Western blot were used to detect the cell F. The expression of protein and mRNA of AP. Results FAP almost did not express in NFs, but the expression of mRNA and protein in FAP increased in CAFs. Compared with group NFs and CAFs, the TGF- beta 1 of 10 ng/ml could obviously promote the secretion of CAFs cells. The expression of FAP increased continuously, the expression of 24 h increased in comparison with 12 h, and the difference was statistically significant (P0.05). Conclusion TGF- beta 1 could promote the expression of FAP in CAFs and play an important role in the epithelial and interstitial interaction of tumor.
