牙胚組織發(fā)育過程中成牙基因的表達(dá)變化

【摘要】:目的:比較DMP1、AMBN、Collagen-Ⅰ和DLX1在SD大鼠牙胚發(fā)育過程中mRNA相對表達(dá)量的差別,探究SD大鼠牙胚分化過程中,這些基因的相對表達(dá)量的變化,揭示階段性表達(dá)模式,探索適合做組織工程牙的種子細(xì)胞。方法:取胚胎14天、17天的SD胎鼠及出生后第4天的SD乳鼠的牙胚組織,并把出生后第4天SD乳鼠的牙胚組織經(jīng)體外培養(yǎng)后得到第1、3、6天的牙胚細(xì)胞均用實(shí)時(shí)定量PCR的方法檢測成牙基因mRNA水平相對表達(dá)量的變化。結(jié)果:牙胚組織中的DMP1、 AMBN、Collagen-Ⅰ和DLX1mRNA相對表達(dá)量在出生后第4天及胚胎17天均高于胚胎14天,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。牙胚細(xì)胞中DMP1、AMBN、Collagen-Ⅰ基因mRNA的相對表達(dá)量在培養(yǎng)第3天均高于培養(yǎng)第1天及第6天,差異有統(tǒng)計(jì)學(xué)意義(P0.05),而DLX1mRNA的相對表達(dá)量在培養(yǎng)第1天均高于培養(yǎng)第3天及第6天,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:出生后第4天的牙胚組織中四個(gè)成牙基因相對表達(dá)量較多可以作為種子細(xì)胞。在培養(yǎng)后第3天的牙胚細(xì)胞是增殖和分化較好的時(shí)期可以用于構(gòu)建組織工程牙。
[Abstract]:Objective: to compare the difference of mRNA relative expression between DMP1 and DLX1 during the development of SD rat tooth germ, to explore the changes of the relative expression of these genes during the development of SD rat tooth germ, and to reveal the expression pattern of these genes. To explore suitable seed cells for tissue engineering teeth. Methods: tooth germs were taken from SD fetal rats of 14 days and 17 days of gestation and SD newborn rats on the 4th day after birth.
