LAIR-1在口腔扁平苔蘚患者中表達(dá)的研究

【摘要】:實(shí)驗(yàn)?zāi)康模和ㄟ^檢測(cè)口腔扁平苔蘚(OLP)患者外周血中LAIR-1及相關(guān)細(xì)胞因子的表達(dá),探討LAIR-1及相關(guān)細(xì)胞因子在口腔扁平苔蘚發(fā)生發(fā)展中的作用,進(jìn)而為OLP發(fā)病機(jī)制提供理論依據(jù),為治療口腔扁平苔蘚提供新的更為有效的方法。實(shí)驗(yàn)方法:收集口腔扁平苔蘚患者22位,采集患者外周血3m1,同時(shí)選取22位健康人作為正常對(duì)照組,兩組年齡和性別相匹配,2m1新鮮血液用于流式細(xì)胞術(shù)以檢測(cè)LAIR-1在CD4+Foxp3+Treg上的比例,1m1血液凍存用于ELISA檢測(cè)TGF-β及IL-10,數(shù)據(jù)資料采用均數(shù)±標(biāo)準(zhǔn)差(x±s)來表示,并對(duì)口腔扁平苔癬組LAIR-1在CD4+Foxp 3+Tre g上的表達(dá)水平和TGF-β的水平作相關(guān)性分析,統(tǒng)計(jì)學(xué)分析采用SPSS13.0軟件處理,兩組間比較采用兩樣本的T檢驗(yàn)。實(shí)驗(yàn)結(jié)果:1、流式細(xì)胞術(shù)的結(jié)果:OLP患者外周血CD4+Foxp3+Treg細(xì)胞LAIR-1的陽性表達(dá)率高于健康人,其差異具有統(tǒng)計(jì)學(xué)意義(PO.05)。2.ELISA檢測(cè)外周血中細(xì)胞因子水平表達(dá)的結(jié)果:口腔扁平苔蘚患者外周血中,TGF-β、IL-10的表達(dá)均高于正常對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。3、OLP患者外周血LA工R-1在CD4+Foxp3+Treg的表達(dá)與TGF-β的相互關(guān)系:OLP患者CD4+LAIR-1+Foxp3+Treg/CD4+T的表達(dá)比例和TGF-β的表達(dá)呈正相關(guān)(r=0.43,P0.05)。實(shí)驗(yàn)結(jié)論:1、OLP患者外周血中LAIR-1在CD4+FoxP3+Treg上的表達(dá)較健康對(duì)照組呈上升趨勢(shì),表明了在發(fā)病的過程中存在免疫調(diào)節(jié)功能紊亂,LAIR-1和調(diào)節(jié)性T細(xì)胞在該病的發(fā)生發(fā)展過程中占據(jù)重要的角色。2、本實(shí)驗(yàn)研究發(fā)現(xiàn)在OLP患者外周血中,TGF-β表達(dá)升高,機(jī)體免疫功能調(diào)節(jié)出現(xiàn)異常,TGF-β在OLP的發(fā)生發(fā)展過程中發(fā)揮了較為重要的作用。3、OLP患者外周血IL-10的水平呈上升趨勢(shì),其抑制了自身反應(yīng)性淋巴細(xì)胞以及巨噬細(xì)胞的活化和擴(kuò)增,從而維持外周免疫耐受。4、口腔扁平苔蘚患者抑制性受體LAIR-1在CD4+Foxp3+Treg上的表達(dá)與TGF-β的表達(dá)水平呈正相關(guān)(r=0.43,P0.05)。這提示LAIR-1可能是Treg細(xì)胞發(fā)揮免疫抑制作用的重要分子,LAIR-1可能通過對(duì)OLP患者Treg細(xì)胞功能的影響而參與了OLP的發(fā)生。
[Abstract]:Objective: to investigate the role of LAIR-1 and related cytokines in the development of oral lichen planus (OLP) by detecting the expression of LAIR-1 and related cytokines in the peripheral blood of patients with oral lichen planus (OLP), and to provide a theoretical basis for the pathogenesis of OLP. To provide a new and more effective method for the treatment of oral lichen planus. Methods: 22 patients with oral lichen planus and 3ml of peripheral blood were collected and 22 healthy persons were selected as control group. Two groups of age and sex matched 2m1 fresh blood were used for flow cytometry to detect the proportion of LAIR-1 on CD4 Foxp3 Treg. 1m1 was used to detect TGF- 尾 and IL-10 by Elisa. The data were expressed as mean 鹵standard deviation (x 鹵s). The correlation between the expression of LAIR-1 on CD4 Foxp3Tre g and the level of TGF- 尾 in oral lichen planus group was analyzed. SPSS 13.0 software was used to deal with the statistical analysis. The positive expression rate of LAIR-1 in CD4 Foxp3 Treg cells in peripheral blood of patients with OLP was higher than that in healthy people. The difference was statistically significant (PO.05) .2.The expression of TGF- 尾 -IL-10 in peripheral blood of patients with oral lichen planus was significantly higher than that of normal controls. 2. The expression of cytokines in peripheral blood of patients with oral lichen planus was higher than that of normal controls. There was a significant correlation between the expression of R-1 in CD4 Foxp3 Treg and TGF- 尾 in the peripheral blood of patients with OLP (P0.05). There was a positive correlation between the expression of TGF- 尾 and the expression of TGF- 尾 in CD4 LAIR-1 Foxp3 / Treg-T / T (P 0.05). Conclusion the expression of LAIR-1 on CD4 FoxP3 Treg in the peripheral blood of the patients with OLP was higher than that of the healthy controls. These results suggest that the immune regulatory dysfunction of LAIR-1 and regulatory T cells play an important role in the development of OLP. This study found that the expression of TGF- 尾 in peripheral blood of OLP patients was higher than that in patients with OLP, and the expression of TGF- 尾 in the peripheral blood of OLP patients was increased. TGF- 尾 plays an important role in the development of OLP. The level of IL-10 in peripheral blood of patients with OLP is on the rise, which inhibits the activation and expansion of autoreactive lymphocytes and macrophages. Thus, the expression of inhibitory receptor LAIR-1 in CD4 Foxp3 Treg was positively correlated with the expression of TGF- 尾 in patients with oral lichen planus. These results suggest that LAIR-1 may play an important role in the immunosuppressive effect of Treg cells and that LAIR-1 may be involved in the development of OLP by affecting the function of Treg cells in OLP patients.
