超聲對(duì)大鼠正畸性牙根吸收的修復(fù)作用及機(jī)制研究

【摘要】:目的:研究超聲對(duì)大鼠正畸性牙根吸收的作用及其對(duì)OPG/RANKL比值和TNF-α表達(dá)的影響。 方法:64只雄性Wistar大鼠,隨機(jī)分為4組,每組16只分別為:空白對(duì)照組、單純加力組、自然修復(fù)組和超聲治療組。除空白對(duì)照組外,其余組建立正畸性牙根吸收模型。用100g初始力值加載于大鼠右側(cè)上頜中切牙與第一磨牙之間,2w后拆除矯治器,并處死單純加力組。超聲治療組接受頻率1.5MHz、強(qiáng)度100MW/cm2的超聲照射2w。每組隨機(jī)取8只,應(yīng)用掃描電鏡觀察牙根表面的微觀改變,并測(cè)量牙根吸收指數(shù);每組其余8只取實(shí)驗(yàn)牙和其臨近牙周組織,制作以實(shí)驗(yàn)牙為中心,矢狀向的組織切片,HE染色光鏡下觀察牙根吸收與修復(fù)的情況;抗酒石酸酸性磷酸酶(TRAP)染色,行陽(yáng)性細(xì)胞計(jì)數(shù);免疫組化檢測(cè)TNF-α、OPG、RANKL的表達(dá),用Imagepro-Plus6.0圖像分析系統(tǒng)進(jìn)行光密度測(cè)量。對(duì)實(shí)驗(yàn)結(jié)果應(yīng)用spss軟件進(jìn)行單因素方差分析。 結(jié)果: 1.與自然修復(fù)組相比,超聲治療組牙根吸收指數(shù)和破牙骨質(zhì)細(xì)胞的數(shù)目減少了,差異有統(tǒng)計(jì)學(xué)意義;超聲治療組可見(jiàn)有大量的新生牙骨質(zhì)形成。 2.超聲治療組TNF-α的表達(dá)下調(diào),,OPG/RANKL的比值升高,與自然修復(fù)組相比差異有統(tǒng)計(jì)學(xué)意義。 結(jié)論:超聲能夠下調(diào)TNF-α的表達(dá),上調(diào)OPG/RANKL的比值,減少了破牙骨質(zhì)細(xì)胞的生成,有利于牙骨質(zhì)向再生的方向發(fā)展。
[Abstract]:Aim: to study the effects of ultrasound on orthodontic root resorption and the expression of OPG / RANKL and TNF- 偽 in rats. Methods Sixty-four male Wistar rats were randomly divided into 4 groups: blank control group, natural repair group and ultrasound treatment group. In addition to blank control group, orthodontic root resorption model was established in other groups. The orthodontic appliance was removed from the right maxillary central incisor and first molar for 2 weeks with the initial force of 100g, and the treatment group was killed. The ultrasound treatment group received ultrasound irradiation with a frequency of 1.5 MHz and a intensity of 100 MW / cm ~ 2 for 2 weeks. Eight teeth in each group were randomly selected. The microcosmic changes of the root surface were observed by scanning electron microscope, and the root resorption index was measured, and the experimental teeth and their adjacent periodontal tissues were taken from the other 8 teeth in each group to make the experimental teeth as the center. Sagittal tissue sections were stained with HE staining to observe root resorption and repair; tartrate-resistant acid phosphatase (TRAP) staining was used to count positive cells; immunohistochemical staining was used to detect the expression of TNF- 偽 OPGG RANKL, and the optical density was measured by imagepro-Plus6.0 image analysis system. The results of the experiment were analyzed by spss software. Results: 1. Compared with the natural repair group, the root resorption index and the number of osteoclast cells in the ultrasonic treatment group decreased, the difference was statistically significant, and a large number of new cementum formation was observed in the ultrasound treatment group. 2. 2. The down-regulation of TNF- 偽 expression in ultrasound treatment group increased the ratio of OPG / RANKL, which was significantly higher than that in natural repair group. Conclusion: ultrasound can down-regulate the expression of TNF- 偽, upregulate the ratio of OPG / RANKL, reduce the formation of osteoclast, and promote the development of cementum regeneration.
