【摘要】：目的:探討慢性牙周炎患者中吸煙個體血清在具核梭桿菌(Fn)侵入KB細胞及在侵入過程中對KB細胞分泌基質(zhì)金屬蛋白酶-1(MMP-1)的作用。方法:提取慢性牙周炎吸煙個體10例(Y組)、慢性牙周炎非吸煙個體10例(N組)、牙周健康非吸煙個體5例(H組)的血清。分別將各組血清200、400、800μl添加至Fn感染KB細胞模型,作用24 h后計數(shù)細菌菌落。用人酶聯(lián)免疫吸附測定試劑盒檢測各組上清液中MMP-1的濃度。用單因素方差分析進行數(shù)據(jù)分析。結(jié)果:3組800μl血清引起Fn侵入KB細胞的侵入率(%)分別為12.59±1.27、8.03±0.075和7.99±0.14(P0.05),KB細胞分泌MMP-1的濃度(μg/L)分別為400.04±21.02、252.57±18.89、262.47±35.29(P0.05)。200μl和400μl血清組與N組、H組比較,Fn侵入率及KB細胞MMP-1分泌量無統(tǒng)計學(xué)差異(P0.05)。結(jié)論:較高劑量吸煙個體血清有利于Fn侵入KB細胞并促進KB細胞分泌MMP-1。
[Abstract]:Objective: to investigate the effect of serum from smoking patients with chronic periodontitis on invasion of KB cells and secretion of matrix metalloproteinase-1 (MMP-1) in KB cells. Methods: the serum samples were extracted from 10 smokers with chronic periodontitis, 10 patients with chronic periodontitis and 5 healthy nonsmokers. Serum 200400800 渭 l was added to the KB cell model infected with FN for 24 h, and the bacterial colonies were counted. The concentration of MMP-1 in supernatant of each group was detected by Elisa kit. Single factor analysis of variance (ANOVA) was used to analyze the data. Results the invasion rate of FN invading KB cells was 12.59 鹵1.270.03 鹵0.075 and 7.99 鹵0.14P0.05KB cells respectively (渭 g / L) in the three groups (渭 g / L) were 400.04 鹵22252.57 鹵18.89262.47 鹵35.29(P0.05).200 渭 l and 400 渭 l, respectively. There was no significant difference in the invasion rate of FN and the MMP-1 secretion of KBP0.05 cells between the two groups. Conclusion: the higher dose of smoking serum is beneficial to the invasion of FN into KB cells and the secretion of MMP-1 by KB cells.
