【摘要】：目的探討飲酒和細胞外超氧化物岐化酶(EC-SOD)、乙醛脫氫酶2(ALDH2)基因多態(tài)性與口腔鱗狀細胞癌患病之間的關系。方法采用病例對照研究的方法,以750例口腔鱗狀細胞癌患者(病例組)及750例非癌對照者(對照組)的外周血白細胞為樣本,采用聚合酶鏈反應(PCR)技術分析EC-SOD和ALDH2基因的多態(tài)性,并分析該基因多態(tài)性與口腔鱗狀細胞癌患病的關系。結果病例組EC-SOD(C/G)和ALDH2變異基因型頻率分別為38.27%、69.47%,對照組則為21.07%、44.40%,二者差異有統(tǒng)計學意義(P0.01)。EC-SOD(C/G)患口腔鱗狀細胞癌的風險顯著增加(OR=2.32),ALDH2變異基因型的患病風險也顯著增加(OR=2.85)�；蛲蛔兊膮f(xié)同分析發(fā)現(xiàn),EC-SOD(C/G)/ALDH2變異基因型在病例組和對照組中的分布頻率分別為30.67%和6.80%,二者差異有統(tǒng)計學意義(P0.01)。EC-SOD(C/G)/ALDH2變異基因型患口腔鱗狀細胞癌的風險顯著增加(OR=8.13)。病例組的飲酒率明顯高于對照組(OR=2.70),EC-SOD(C/G)及ALDH2變異基因型與飲酒有協(xié)同作用(OR=25.00)。結論 ECSOD及ALDH2變異基因型和飲酒是口腔鱗狀細胞癌的易患因素,三者聯(lián)合在口腔鱗狀細胞癌的發(fā)生中有協(xié)同作用。
[Abstract]:Objective to investigate the relationship between alcohol consumption, extracellular superoxide dismutase (EC-SOD), acetaldehyde dehydrogenase (ALDH2) gene polymorphism and oral squamous cell carcinoma (OSCC). Methods Peripheral blood leukocytes were collected from 750 patients with oral squamous cell carcinoma (SCC) and 750 non-cancerous controls (control group) by a case-control study. Polymerase chain reaction (PCR) technique was used to analyze the polymorphism of EC-SOD and ALDH2 gene, and the relationship between the polymorphism and the prevalence of oral squamous cell carcinoma. Results the genotype frequencies of EC-SODN C / G) and ALDH2 were 38.27 and 21.07 respectively in the case group and 44.40 in the control group. There was a significant difference between the two groups in the risk of oral squamous cell carcinoma (OSCC). The risk of OR2.32ALDH2 variant genotype was also significantly increased. The co-analysis of gene mutation showed that the distribution frequency of the genotype of EC-SODG / ALDH2 mutation was 30.67% and 6.80 in the case group and the control group, respectively. There was a significant difference between the two genotypes. The risk of oral squamous cell carcinoma was significantly increased by the genotype of the C- SODX / C / Gr / ALDH2 mutation in the patients with oral squamous cell carcinoma (OSCC), and the risk of oral squamous cell carcinoma (OSCC) was significantly higher than that in the control group (P < 0.05). The drinking rate in the case group was significantly higher than that in the control group (2.70 EC-SOD C / G) and the genotype of ALDH2 mutation had a synergistic effect on alcohol consumption. Conclusion ECSOD and ALDH2 genotypes and alcohol consumption are the predisposing factors of oral squamous cell carcinoma, and there is a synergistic effect among them in the occurrence of oral squamous cell carcinoma.
