統(tǒng)計(jì)方法在研究口腔微生物菌群群落多樣性維持機(jī)制中的應(yīng)用

【摘要】:了解微生物群落對人類的健康作用是新興生物醫(yī)學(xué)的重大挑戰(zhàn),而群落里的結(jié)構(gòu)和功能又是維持生態(tài)平衡的關(guān)鍵。我們通過一系列的模型分析,旨在揭示人體微生物群落的結(jié)構(gòu)以及物種共存機(jī)制,對后續(xù)深入研究與牙周炎相關(guān)的微生物群落的結(jié)構(gòu)、功能及其穩(wěn)定性有著重要的理論及實(shí)踐意義。本文研究的對象是牙周炎患者口腔微生物,牙周炎是最為常見的口腔疾病之一,已有大量研究表明牙周炎的發(fā)生與口腔微生物群落的失衡有關(guān),因此本論文從生態(tài)學(xué)角度利用統(tǒng)計(jì)方法研究牙周炎有關(guān)的口腔微生物群落的維持機(jī)制。首先,用等級多度曲線和物種多度分布曲線基礎(chǔ)的描述群落組成結(jié)構(gòu),其次用Ewens的抽樣模型和Etienne的抽樣模型公式檢測口腔微生物群落是否符合中性理論,以此研究牙周炎對口腔微生物菌群結(jié)構(gòu)和功能的影響;最后,用Taylor冪法則和擴(kuò)展的Taylor冪法則研究口腔微生物群落的空間的異質(zhì)性,以探討牙周炎是否與微生物群落的空間異質(zhì)性的改變有關(guān)。我們通過實(shí)例數(shù)據(jù)的模型分析,對于觀察到的口腔微生物群落而言,兩個抽樣模型證實(shí)了在口腔微生物群落中傳播限制并沒有發(fā)揮什么作用,也發(fā)現(xiàn)了牙周炎患者的微生物群落顯示生物多樣性高于健康人,發(fā)現(xiàn)生物群落的結(jié)構(gòu)影響著生態(tài)的平衡,Taylor冪法則和擴(kuò)展的Taylor冪法模型結(jié)果說明,牙周炎患者口腔的生態(tài)環(huán)境較健康對照發(fā)生了 一定的改變,使得微生物的種間差異增大,這種環(huán)境的改變可能是非隨機(jī)的,因而使得種內(nèi)異質(zhì)性沒有發(fā)生明顯改變,總之牙周炎的發(fā)生可能與口腔微生態(tài)環(huán)境的破壞有著直接關(guān)聯(lián)。
[Abstract]:Understanding the effects of microbial communities on human health is a major challenge in emerging biomedicine, and the structure and function of the community is the key to maintain ecological balance. Through a series of model analysis, we aim to reveal the structure of human microbial community and the mechanism of species coexistence, and further study the structure of microbial community related to periodontitis. Function and its stability have important theoretical and practical significance. The subjects of this study are oral microbes in patients with periodontitis. Periodontitis is one of the most common oral diseases. A large number of studies have shown that the occurrence of periodontitis is related to the imbalance of oral microbial community. In this paper, the maintenance mechanism of oral microbial community related to periodontitis was studied by using statistical method from the point of view of ecology. Firstly, community composition was described on the basis of grade abundance curve and species abundance distribution curve. Secondly, Ewens's sampling model and Etienne's sampling model formula were used to detect whether oral microbial community was in accordance with neutral theory. The effects of periodontitis on the structure and function of oral microflora were studied. Finally, the spatial heterogeneity of oral microbial community was studied by Taylor power law and extended Taylor power law. To explore whether periodontitis is related to the change of spatial heterogeneity of microbial community. Through the model analysis of the sample data, for the observed oral microbe community, two sampling models confirmed that the transmission limitation in the oral microbe community did not play a significant role. It was also found that the microbial community of periodontitis patients showed higher biodiversity than that of healthy people, and the balance Taylor's power law and the extended Taylor power method model showed that the structure of the biological community affected the ecology. The oral ecological environment of periodontitis patients was changed to a certain extent, which increased the difference of microbes between species, which may be non-random, so that the intraspecific heterogeneity did not change significantly. In short, periodontitis may be directly related to oral microecological environment damage.
