【摘要】:目的: 口腔鱗狀細胞癌(oral squamous cell carcinima, OSCC)惡性程度高,浸潤性強,且易發(fā)生淋巴結(jié)及遠處臟器轉(zhuǎn)移。目前針對OSCC的研究較多的集中于細胞凋亡和增殖的平衡機制方面,Livin和Caspase-3是細胞凋亡和增殖過程中重要的生物學指標。本實驗通過探討凋亡抑制蛋白(IAP) Livin和半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)在口腔鱗狀細胞癌(OSCC)中的表達及其相關性,從分子學水平研究Livin和Caspase-3mRNA在口腔鱗狀細胞癌(OSCC)和正常口腔黏膜組織(NOT)中的表達,分析二者在口腔鱗狀細胞癌中表達的相關性,為對患者診斷及監(jiān)測、評估術(shù)后復發(fā)、轉(zhuǎn)移的風險程度提供參考,以輔助臨床工作。 方法: 本研究應用逆轉(zhuǎn)錄-聚合酶鏈反應(reverse transcription polymerase chain reaction, RT-PCR)方法,選取2011年10月~2013年2月在安徽醫(yī)科大學第一附屬醫(yī)院口腔頜面外科就診,并均經(jīng)組織病理檢查確診為OSCC患者新鮮組織標本45例,所有OSCC組患者均為首次確診,術(shù)前均未行放、化療或其他抗腫瘤治療,均無遠端臟器的轉(zhuǎn)移,無其他惡性腫瘤史,無家族腫瘤史。另選11例患者為陰性對照組(NOT組),其中7例取自阻生齒拔除患者,4例取自牙槽嵴修整患者。所有標本均于術(shù)中取材后立即置于液氮中,然后移至-80℃超低溫冰箱中貯存?zhèn)溆谩?結(jié)果: 1.Livin mRNA和Caspase-3mRNA在OSCC組織中的相對含量均與組織分化程度、淋巴結(jié)轉(zhuǎn)移和臨床分期均有相關性;但與患者的年齡、性別無關。 2.Livin和Caspase-3在45例OSCC和11例NOT中均有不同程度的表達:Caspase-3mRNA在OSCC組織中的陽性表達率明顯低于NOT (P0.05);其在NOT中相對含量(0.193±0.031),高于OSCC(0.102±0.025)(P0.01)。Livin mRNA在OSCC組織中的陽性表達率明顯高于NOT (P0.05);其表達量在OSCC組織中為(0.542±0.074),明顯高于NOT (0.181±0.026)(P0.01). 3.根據(jù)Pearson列聯(lián)系數(shù)相關分析, Livin mRNA和Caspase-3mRNA在OSCC組織中的表達呈相關關系(P0.01)。 結(jié)論: Livin和Caspase-3在OSCC中的表達結(jié)果分析顯示,兩者可能參與了OSCC的發(fā)生、發(fā)展的過程,聯(lián)合檢測Livin與Caspase-3可以為研究OSCC發(fā)病機制提供新的思路,為OSCC的臨床分級、分期、評估患者預后提供數(shù)據(jù)支持。
[Abstract]:Objective: Oral squamous cell carcinima, OSCC) in oral squamous cell carcinoma is highly malignant, invasive and prone to lymph node and distant organ metastasis. At present, the researches on OSCC focus on the balance mechanism of apoptosis and proliferation. Livin and Caspase-3 are important biological indexes in the process of cell apoptosis and proliferation. In this study, we investigated the expression of Livin and caspase-3 in oral squamous cell carcinoma (OSCC) and their correlation with the expression of apoptosis-inhibiting protein (IAP) and cysteine aspartate proteinase-3 (Caspase-3) in oral squamous cell carcinoma (OSCC). To study the expression of Livin and Caspase-3mRNA in oral squamous cell carcinoma (OSCC) and normal oral mucosa tissue from molecular level, and to analyze the correlation between them in oral squamous cell carcinoma (OSCC) and normal oral mucosa. The degree of risk of transfer provides a reference to assist clinical work. Methods: In this study, reverse transcription polymerase chain reaction, RT-PCR method was used to select oral and maxillofacial surgery patients from October 2011 to February 2013 in the first affiliated Hospital of Anhui Medical University. 45 cases of fresh tissue specimens were confirmed by histopathological examination. All the patients in OSCC group were diagnosed for the first time. No radiotherapy, chemotherapy or other antitumor therapy, no metastasis of distal organs and no history of other malignant tumors were performed before operation. No family history of cancer. Another 11 cases were selected as negative control group, 7 cases were extracted from impacted teeth, 4 cases were from alveolar ridge dressing.
