丹參對(duì)大鼠正畸牙牙周組織中OPG RANKL表達(dá)的影響

【摘要】:目的:研究丹參注射液對(duì)大鼠正畸牙壓力側(cè)牙周膜中RANKL和OPG表達(dá)的影響。 方法:清潔級(jí)SD雄性大鼠240只,隨機(jī)分為A正畸加力組,B丹參干預(yù)組,C丹參對(duì)照組,D空白對(duì)照組,AD兩組與BC兩組每天于左上頜第一磨牙頰側(cè)黏膜下分別注射0.1mlPBS或者丹參注射液;在實(shí)驗(yàn)開始后第l、3、5、7、10、14天分批處死,測量左上頜第一磨牙近中移動(dòng)距離,免疫組織化學(xué)檢測RANKL和OPG的表達(dá),TRAP染色檢測破骨細(xì)胞。 結(jié)果:丹參干預(yù)組自實(shí)驗(yàn)第5天起牙齒移動(dòng)距離顯著大于正畸加力組(P<0.05);正畸加力組與丹參干預(yù)組之間RANKL表達(dá)無明顯差異(P>0.05);丹參干預(yù)組自實(shí)驗(yàn)第7天起OPG的表達(dá)顯著大于正畸加力組(P<0.05);在實(shí)驗(yàn)第3天和第5天,,丹參干預(yù)組中的破骨細(xì)胞數(shù)目顯著大于正畸加力組(P<0.05)。 結(jié)論:丹參注射液對(duì)大鼠正畸牙壓力側(cè)的RANKL表達(dá)無明顯影響,卻可以使壓力側(cè)出現(xiàn)更多的破骨細(xì)胞從而加速牙周改建;同時(shí)能夠促使大鼠正畸牙壓力側(cè)的OPG表達(dá)上調(diào),但不能將OPG到達(dá)峰值的時(shí)間提前,使其在加速正畸牙移動(dòng)的同時(shí)維持骨量。
[Abstract]:Aim: to study the effect of Salvia miltiorrhiza injection on the expression of RANKL and OPG in orthodontic periodontal ligament of orthodontic teeth. Methods: 240 male SD rats of clean grade were randomly divided into two groups: group A, orthodontic treatment group, group B, group C, the control group of Salvia miltiorrhiza, group C, the control group D, the control group, AD and group BC, respectively, 0.1mlPBS or salvia miltiorrhiza injection were injected into the buccal mucosa of the left maxillary first molar every day. At the beginning of the experiment, the first molar of left maxillary was killed in batches on the 14th day. The distance of proximal movement of left maxillary first molar was measured. The expression of RANKL and OPG was detected by trap staining. The osteoclasts were detected by immunohistochemistry. Results: the distance of tooth movement in Danshen intervention group was significantly higher than that in orthodontic plus force group (P < 0.05), but there was no significant difference in RANKL expression between orthodontic and salvia miltiorrhiza intervention group (P > 0.05), and the expression of OPG in salvia miltiorrhiza intervention group from the 7th day of experiment was significantly higher than that of orthodontic treatment group (P < 0.05). It was larger than that in orthodontic stress group (P < 0.05), and on the 3rd and 5th day, The number of osteoclasts in salvia miltiorrhiza intervention group was significantly higher than that in orthodontic treatment group (P < 0.05). Conclusion: Salvia miltiorrhiza injection has no obvious effect on the expression of RANKL on the pressure side of orthodontic teeth, but it can make more osteoclasts appear in the pressure side of the orthodontic teeth and accelerate the periodontal remodeling. It can also promote the expression of OPG in the pressure side of orthodontic teeth. However, the peak time of OPG can not be brought forward, so that the bone mass can be maintained while accelerating orthodontic tooth movement.
