【摘要】：目的:探討Wnt通路抑制因子Dickkopf1(DKK1)在出生后小鼠牙發(fā)育各階段的時(shí)空表達(dá)特點(diǎn),為闡明Wnt信號(hào)通路調(diào)控牙發(fā)育機(jī)制提供理論依據(jù)。方法:選擇出生后第0.5、6.5、12.5、18.5、24.5和30.5天的昆明小鼠并按時(shí)間分組,每組3只。頸椎脫臼法處死小鼠,分離包含第一磨牙的下頜骨組織后進(jìn)行固定、脫鈣處理,制作厚度為5μm的下頜第一磨牙牙體-牙周組織聯(lián)合切片。采用蘇木素-伊紅(HE)染色觀察下頜第一磨牙的發(fā)育狀態(tài),應(yīng)用免疫組織化學(xué)染色方法觀察小鼠牙體組織和牙周組織中DKK1的表達(dá)特點(diǎn)。結(jié)果:小鼠出生后第0.5天時(shí),下頜第一磨牙牙胚處于鐘狀晚期;第6.5天時(shí),下頜第一磨牙牙釉質(zhì)發(fā)育基本完成,可見(jiàn)上皮根鞘向根方延伸;第12.5天時(shí),冠部牙本質(zhì)發(fā)育完成,牙根形成約1/3;第18.5天時(shí),牙根形成約2/3;第24.5天時(shí)牙根發(fā)育達(dá)到全長(zhǎng);第30.5天時(shí)根尖孔縮窄,牙根發(fā)育基本完成。小鼠出生后第0.5天時(shí),牙胚中無(wú)DKK1陽(yáng)性表達(dá);第6.5天時(shí),成牙本質(zhì)細(xì)胞中DKK1呈弱表達(dá);從第12.5天到30.5天,成牙本質(zhì)細(xì)胞中DKK1仍呈陽(yáng)性表達(dá),而牙周膜中開(kāi)始出現(xiàn)DKK1表達(dá),且隨時(shí)間延長(zhǎng)表達(dá)逐漸增強(qiáng),牙槽骨和細(xì)胞性牙骨質(zhì)中也出現(xiàn)DKK1的陽(yáng)性表達(dá),牙髓組織中始終呈陰性表達(dá)。結(jié)論:出生后小鼠牙不同發(fā)育期DKK1呈規(guī)律性表達(dá),提示其可能參與了牙本質(zhì)和牙周組織的發(fā)育過(guò)程。
[Abstract]:Aim: to investigate the temporal and spatial expression of Wnt pathway inhibitor Dickkopf1 DKK1 in postnatal mouse tooth development, and to provide theoretical basis for elucidating the mechanism of tooth development regulated by Wnt signaling pathway. Methods: Kunming mice were divided into three groups according to their time. The mice were killed by cervical dislocations, the mandibular tissues containing the first molars were separated and fixed, decalcified, and the mandibular first molars with thickness of 5 渭 m were made into the joint section of the mandibular first molar and periodontal tissue. The developmental state of mandibular first molar was observed by hematoxylin-eosin-hehe staining and the expression of DKK1 in mouse tooth and periodontal tissue was observed by immunohistochemical staining. Results: on the 0.5 day after birth, the first mandibular molar tooth germ was in the late bell shape, at 6.5 days, the enamel development of the first mandibular molar was basically completed, and the epithelial root sheath extended to the root, and on the 12.5 day, the development of the dentin of the crown was completed. Root formation was about 1 / 3; at 18.5 days, root formation was about 2 / 3; at 24.5 days, the root development reached full length; at 30.5 day, the apical foramen narrowed and root development was basically completed. There was no positive expression of DKK1 in dental germ at 0.5 day after birth, weak expression of DKK1 in odontoblast at 6.5 day, DKK1 expression in odontoblast from 12.5 day to 30.5 day, and DKK1 expression in periodontal membrane. The positive expression of DKK1 was also found in alveolar bone and cellular cementum, but negative in pulp tissue. Conclusion: the expression of DKK1 in different developmental stages of postnatal mouse teeth is regular, which suggests that DKK1 may be involved in the development of dentin and periodontal tissue.
