【摘要】：目的:研究wnt1在口腔黏膜下纖維性變(OSF)患者治療前后的表達(dá)并探討其在OSF發(fā)生發(fā)展中的作用。方法:40例OSF中期患者采用曲安奈德+丹參局部封閉治療4周,治療前及第5周復(fù)診時(shí)記錄VAS及張口度,取唾液及齦溝液ELISA檢測(cè)wnt1表達(dá),實(shí)時(shí)熒光定量PCR檢測(cè)OSF病變頰黏膜中wnt1 mRNA表達(dá)。結(jié)果:治療前OSF組wnt1表達(dá)[頰黏膜定量PCR(36.89±10.40)×10~(-5),唾液濃度(61.61±4.45)ng/L,齦溝液濃度(56.20±3.65)ng/L]均高于正常組[頰黏膜定量PCR(4.63±1.53)×10~(-5),唾液濃度(40.26±3.00)ng/L,齦溝液濃度(53.45±1.74)ng/L](P0.01);OSF組經(jīng)治療后,唾液與齦溝液中wnt1表達(dá)均降低,且與OSF臨床嚴(yán)重程度呈正相關(guān)(P0.05)。結(jié)論:wnt1可能參與OSF的發(fā)生發(fā)展,其在唾液與齦溝液中的實(shí)時(shí)監(jiān)測(cè)有望成為評(píng)價(jià)OSF診療的無(wú)創(chuàng)性檢控手段之一。
[Abstract]:Objective: to investigate the expression of wnt1 in patients with oral submucous fibrosis before and after treatment and its role in the development of OSF. Methods 40 middle stage OSF patients were treated with triamcinolone acetonide salvia miltiorrhiza for 4 weeks. VAS and mouth opening were recorded before and 5 weeks after treatment. Saliva and gingival crevicular fluid ELISA were taken to detect the expression of wnt1. The expression of wnt1 mRNA in buccal mucosa of OSF was detected by real-time fluorescence quantitative PCR. Results: before treatment, the expression of wnt1 [buccal mucosal quantitative PCR(36.89 鹵10.40) 脳 10 ~ (-5) ng / L, saliva concentration (61.61 鹵4.45 ng / L), gingival crevicular fluid concentration (56.20 鹵3.65)ng/L)] in OSF group was higher than that in normal group [buccal mucosa quantitative PCR(4.63 鹵1.53) 脳 10 ~ (-5) ~ (-5), saliva concentration was 40.26 鹵3.00 ng / L, gingival crevicular fluid concentration was 53.45 鹵1.74)ng/L] after treatment, the expression of wnt1 in saliva and gingival crevicular fluid decreased in OSF group. There was a positive correlation with the clinical severity of OSF (P 0.05). ConclusionWnt1 may be involved in the development of OSF and its real-time monitoring in saliva and gingival crevicular fluid is expected to be one of the non-invasive methods to evaluate the diagnosis and treatment of OSF.
