【摘要】：目的:通過建立自體骨髓間充質(zhì)干細(xì)胞復(fù)合富血小板纖維蛋白修復(fù)兔拔牙窩骨缺損的動(dòng)物模型,探討Notch1和Wnt3a信號(hào)通路分子在牙槽骨缺損修復(fù)過程中的表達(dá)及意義。材料及方法:選用36只健康雄性2月齡新西蘭兔,隨機(jī)分為四組,全麻下均微創(chuàng)拔除下頜左側(cè)中切牙。A組植入自體骨髓間充質(zhì)干細(xì)胞與富血小板纖維蛋白復(fù)合物,B、C組分別植入單一富血小板纖維蛋白、自體骨髓間充質(zhì)干細(xì)胞,D組無任何材料植入的空白對(duì)照。術(shù)后4、8、12周(每個(gè)時(shí)間點(diǎn)3只)處死動(dòng)物并立即在骨缺損同一部位取材,制作骨標(biāo)本。利用免疫組化和免疫熒光檢測(cè)骨缺損修復(fù)過程中Notch1和Wnt3a的表達(dá)情況。結(jié)果:免疫組化顯示第4和8周A、B、C組Notch1和Wnt3a表達(dá)均大于D組(P0.05)。12周時(shí),D組這兩個(gè)通路信號(hào)分子的表達(dá)大于其他三組(P0.05)。免疫熒光說明在骨缺損區(qū)新生骨細(xì)胞的Notch1和Wnt3a表達(dá)第4周時(shí)達(dá)到高峰,隨著時(shí)間的延長(zhǎng)骨缺損修復(fù)完成,表達(dá)消失。結(jié)論:Notch1和Wnt3a信號(hào)分子在自體骨髓間充質(zhì)干細(xì)胞復(fù)合富血小板纖維蛋白促進(jìn)骨缺損修復(fù)過程中表達(dá),它們通過調(diào)控自體骨髓間充質(zhì)干細(xì)胞的增殖與分化加速牙槽骨缺損修復(fù)的愈合過程。并且兩者表達(dá)相似,有可能存在“串話”。
[Abstract]:Objective: to investigate the expression and significance of Notch1 and Wnt3a signal pathway molecules in alveolar bone defect repair by establishing an animal model of autogenous bone marrow mesenchymal stem cells combined with platelet-rich fibrin. Materials and methods: 36 healthy male 2-month-old New Zealand rabbits were randomly divided into four groups. The left central incisor of the mandible was extracted by minimally invasive extraction under general anesthesia. Group A was implanted with autologous bone marrow mesenchymal stem cells and platelet-rich fibrin complex. Group C was implanted with a single platelet-rich fibrin. Autologous bone marrow mesenchymal stem cells group D without any material implantation blank control. The animals were killed at 4 weeks and 8 weeks after operation (3 at each time point) and bone specimens were made from the same site of bone defect immediately. Immunohistochemistry and immunofluorescence were used to detect the expression of Notch1 and Wnt3a during bone defect repair. Results: the expression of Notch1 and Wnt3a in group C was higher than that in group D at the 4th and 8th week. The expression of signal molecules of these two pathways in group D was higher than that in group D at week 12. Immunofluorescence showed that the expression of Notch1 and Wnt3a reached its peak at the 4th week in the new bone cells in the bone defect area, and the expression disappeared with the completion of the repair of the bone defect. Conclusion the expression of Wnt3a signaling molecules in autologous bone marrow mesenchymal stem cells combined with platelet-rich fibrin promotes the repair of bone defect. They accelerate the healing of alveolar bone defects by regulating the proliferation and differentiation of autologous bone marrow mesenchymal stem cells. And the two expressions are similar, there may be a "crosstalk".
