【摘要】：目的:觀察NF-κB非經(jīng)典通路IKKα調(diào)控的Maspin在HPDLCs中的表達(dá)。方法:組織塊法原代培養(yǎng)HPDLCs。倒置顯微鏡及SP免疫組化鑒定細(xì)胞來(lái)源。免疫細(xì)胞化學(xué)定位Maspin在HPDLCs中的表達(dá)。實(shí)驗(yàn)分空白對(duì)照組(NC)、LPS_(1-4)組(LPS終濃度為0.1、1、10、50mg/L)。RT-PCR及Western Blot分別檢測(cè)LPS刺激下HPDLCs中Maspin、IKKαmRNA及蛋白的表達(dá)。Elisa檢測(cè)炎性因子TNF-α和IL-1β的分泌。結(jié)果:Maspin在正常與LPS刺激的HPDLCs中均呈胞漿表達(dá)。隨著LPS濃度增加Maspin蛋白表達(dá)下調(diào),LPS_(1-4)與NC組相比(P0.05);IKKα蛋白表達(dá)上調(diào),LPS_(1-4)與NC組相比(P0.05)。mRNA表達(dá)與蛋白表達(dá)趨勢(shì)相同。同時(shí),二者相關(guān)性分析可知,Maspin與IKKαmRNA及蛋白表達(dá)呈負(fù)相關(guān)。Elisa結(jié)果顯示TNF-α、IL-1β隨著LPS濃度增加分泌增加,且與Maspin蛋白表達(dá)呈負(fù)相關(guān)。結(jié)論:Maspin在HPDLCs中呈胞漿表達(dá),LPS可介導(dǎo)HPDLCs中Maspin表達(dá)下調(diào),與TNF-α、IL-1β呈負(fù)相關(guān),且Maspin表達(dá)下調(diào)可能經(jīng)由NF-κB非經(jīng)典途徑IKKα調(diào)控。
[Abstract]:Aim: to investigate the expression of Maspin regulated by NF- 魏 B nonclassical pathway IKK 偽 in HPDLCs. Methods: HPDLCs were cultured by tissue block method. The origin of the cells was identified by inverted microscope and SP immunohistochemistry. Immunocytochemical localization of Maspin expression in HPDLCs. Western Blot and RT-PCR were used to detect the expression of Maspin Ike 偽 mRNA and protein in HPDLCs stimulated by LPS. Elisa was used to detect the secretion of inflammatory factors TNF- 偽 and IL-1 尾. Results both normal and LPS stimulated HPDLCs showed cytoplasmic expression of 1% Maspin. With the increase of LPS concentration, the expression of Maspin protein was down-regulated. (compared with NC group, the expression of P0.05Ik 偽 protein was up-regulated.) compared with NC group, the expression of Maspin protein was similar to that of NC group. Meanwhile, Maspin was negatively correlated with the expression of IKK 偽 mRNA and protein. Elisa results showed that the secretion of TNF- 偽 and IL-1 尾 increased with the increase of LPS concentration, and negatively correlated with the expression of Maspin protein. Conclusion the cytosolic expression of Maspin in HPDLCs can mediate the down-regulation of Maspin expression in HPDLCs, which is negatively correlated with the expression of TNF- 偽 and IL-1 尾, and the down-regulation of Maspin expression may be regulated by IKK 偽 via NF- 魏 B non-classical pathway.
