【摘要】：目的:研究血鏈球菌細(xì)菌素(簡(jiǎn)稱血鏈素)對(duì)牙齦卟啉單胞菌(Porphyromonas gingivalis,P.g)形貌特征及力學(xué)性質(zhì)的影響。方法:1)復(fù)蘇、傳代并鑒定血鏈球菌標(biāo)準(zhǔn)菌株ATCC10556、牙齦卟啉單胞菌標(biāo)準(zhǔn)菌株ATCC33277。2)將血鏈球菌低溫高速離心、細(xì)胞破碎等方法提取血鏈素,使之作用于P.g,于37℃厭氧培養(yǎng)48h。3)采用原子力顯微鏡(AFM)的接觸模式,觀察血鏈素作用前后P.g菌體的形貌變化。4)利用AFM測(cè)定血鏈素作用前后P.g的力-距離曲線,并計(jì)算黏附力及楊氏模量。結(jié)果:1)通過(guò)AFM的觀察:P.g在血鏈素的作用下,菌體直徑縮小、平均粗糙度及平均峰高度增加(P0.05)。2)通過(guò)AFM的觀察:血鏈素作用后,P.g的黏附力、楊氏模量顯著降低(P0.001)。正常P.g的黏附力平均值為(0.70±0.10)nN、楊氏模量平均值為(5.54±0.16)MPa;經(jīng)血鏈素作用后P.g的黏附力平均值為(0.50±0.10)nN、楊氏模量平均值為(3.97±0.64)MPa。結(jié)論:1)血鏈素可引起牙齦卟啉單胞菌形貌特征發(fā)生改變。2)血鏈素可導(dǎo)致牙齦卟啉單胞菌的黏附力及楊氏模量降低。
[Abstract]:Aim: to study the effect of streptococcus sanguis bacteriocin on the morphology and mechanical properties of Porphyromonas gingivalis P.g. Methods: resuscitation, subculture and identification of Streptococcus sanguis standard strain ATCC 10556, standard strain ATCC 33277.2) the Streptococcus sanguis was centrifuged at low temperature and high speed, and its blood chain was extracted by cell fragmentation.
