阿司匹林對兔頰VX-2鱗狀細胞癌炎癥微環(huán)境中樹突細胞的影響

【摘要】:目的探索阿司匹林及炎癥對兔頰VX-2鱗狀細胞癌上清液中樹突細胞(DC)成熟及功能的影響。方法通過瘤粒植入術及機械創(chuàng)傷+高糖飲食的方法,建立兔頰VX-2鱗狀細胞癌及其炎癥模型,將模型兔分為3組。實驗組:制造頰癌及炎癥后使用阿司匹林灌胃治療3 d;對照組:制造頰癌及炎癥后以生理鹽水灌胃作為對照;空白組:不制造炎癥的單純腫瘤兔組。3 d后收集各組腫瘤標本,制成勻漿,取上清液與正常兔外周血單核細胞共培養(yǎng)以使其向DC分化,采用流式細胞儀檢測DC表面分子CD83、CD86、人類白細胞DR抗原(HLA-DR)的表達,混合淋巴細胞反應檢測DC刺激T細胞增殖的能力。結果實驗組和對照組中CD83、CD86、HLA-DR陽性率均低于空白組(P0.05),而實驗組和對照組間的差異無統(tǒng)計學意義(P0.05);同樣,實驗組和對照組刺激T細胞增殖的能力弱于空白組(P0.05),而兩組間刺激T細胞增殖的能力無明顯差異(P0.05)。結論炎癥可抑制DC表面分子CD83、CD86、HLA-DR的表達和功能發(fā)揮,短期、小劑量服用阿司匹林對兔頰VX-2鱗狀細胞癌炎癥微環(huán)境中DC的表型改善和功能發(fā)揮無明顯作用。
[Abstract]:Objective to investigate the effects of aspirin and inflammation on the maturation and function of dendritic cells in the supernatant of rabbit buccal VX-2 squamous cell carcinoma.Methods the rabbit model of VX-2 squamous cell carcinoma and its inflammation was established by means of tumor granulation implantation and high glucose diet after mechanical trauma. The model rabbits were divided into 3 groups.Experimental group: buccal carcinoma and inflammation were treated with aspirin for 3 days; control group: buccal carcinoma and inflammation were treated with normal saline as control group; blank group: rabbit group without inflammation was treated with aspirin for 3 days.The supernatant was cocultured with normal rabbit peripheral blood monocytes to differentiate into DC. Flow cytometry was used to detect the expression of CD83 CD86, HLA-DR antigen of human leukocyte.The ability of DC to stimulate T cell proliferation was detected by mixed lymphocyte reaction.Results the positive rate of HLA-DR in the experimental group and the control group was lower than that in the blank group, but the difference between the experimental group and the control group was not statistically significant (P 0.05).The ability of stimulating T cell proliferation in the experimental group and the control group was weaker than that in the blank group, but there was no significant difference between the two groups in stimulating T cell proliferation.Conclusion inflammation can inhibit the expression and function of HLA-DR on DC surface molecule CD83 and CD86. In short term, low dose aspirin has no effect on the phenotypic improvement and function of DC in the inflammatory microenvironment of rabbit buccal VX-2 squamous cell carcinoma.
