細(xì)辛揮發(fā)油提取及對(duì)牙周炎主要致病菌的作用

【摘要】:牙周炎是一種侵犯牙周支持組織的慢性感染性疾病,是成人牙齒喪失的首要原因,其主要由齦下的具核梭桿菌、中間普氏菌和牙齦卟啉單胞菌等厭氧菌引起。目前針對(duì)利用天然植物高效、安全地防治牙周炎的報(bào)道較少,而細(xì)辛是我國(guó)的一種傳統(tǒng)中草藥,藥理作用廣泛,具有鎮(zhèn)痛、止咳、抗炎、抗過(guò)敏、抗氧化等作用。 本研究采用單因素試驗(yàn)考察不同液料比、蒸餾時(shí)間、顆粒直徑和浸泡時(shí)間對(duì)揮發(fā)油提取率的影響,通過(guò)L9(3)3正交試驗(yàn)確定揮發(fā)油提取的最佳工藝。結(jié)果表明,對(duì)揮發(fā)油提取率影響作用大小為液料比蒸餾時(shí)間顆粒直徑。最優(yōu)水平組合為液料比17、顆粒直徑2.5D95≤3.8mm和蒸餾時(shí)間2h。應(yīng)用氣相色譜-質(zhì)譜聯(lián)用儀(GC-MS)分析細(xì)辛揮發(fā)油單體組分,共鑒定出31種成分,主要為甲基丁香酚、3,5-二甲氧基甲苯和黃樟醚。 采用液體二倍稀釋法和平板涂布法測(cè)定細(xì)辛揮發(fā)油對(duì)牙周炎主要致病菌(具核梭桿菌、中間普氏菌、牙齦卟啉單胞菌)及血鏈球菌的最小抑菌濃度和最小殺菌濃度。結(jié)果表明,細(xì)辛揮發(fā)油對(duì)具核梭桿菌、中間普氏菌、牙齦卟啉單胞菌和血鏈球菌的最小抑菌濃度分別為0.01%、0.04%、0.005%和0.04%,最小殺菌濃度分別為0.02%、0.08%、0.005%和0.32%。評(píng)價(jià)細(xì)辛揮發(fā)油對(duì)小鼠牙槽骨吸收的抑制作用,管飼100μL含2%羧甲基纖維素鈉具核梭桿菌的菌懸液(109CFU/mL)于小鼠口腔,每隔2天1次,共3次,以此建立小鼠牙周炎模型。給予實(shí)驗(yàn)組小鼠2%細(xì)辛揮發(fā)油100μL,陰性對(duì)照組小鼠2%低粘度的羧甲基纖維素鈉,每隔3天1次,持續(xù)6周;空白對(duì)照組小鼠一直飲用無(wú)菌水,不做其他處理。結(jié)果表明,陰性對(duì)照組與空白對(duì)照組相比,小鼠釉質(zhì)骨質(zhì)界到牙槽嵴頂(CEJ-ABC)的距離明顯增大,牙槽骨吸收呈極顯著差異(P0.01);實(shí)驗(yàn)組與陰性對(duì)照組相比,小鼠釉質(zhì)骨質(zhì)界到牙槽嵴頂?shù)木嚯x減小,牙槽骨吸收顯著下降(P0.05)。 評(píng)價(jià)細(xì)辛揮發(fā)油在體內(nèi)的安全性,給予小鼠20%細(xì)辛揮發(fā)油100μL,每天1次,持續(xù)20天,制作小鼠肝臟、腎臟組織的病理切片,經(jīng)蘇木精-伊紅(HE)染色,結(jié)果表明,揮發(fā)油實(shí)驗(yàn)組和空白對(duì)照組未見(jiàn)明顯不同。 綜上所述,細(xì)辛揮發(fā)油對(duì)作為牙周炎主要致病菌的具核梭桿菌、中間普氏菌和牙齦卟啉單胞菌具有明顯的抗菌活性,且治療由具核梭桿菌所致的小鼠實(shí)驗(yàn)性牙周炎效果良好,細(xì)辛揮發(fā)油有望成為一種治療牙周炎的新型天然藥物。
[Abstract]:Periodontitis is a chronic infectious disease that invades periodontal supporting tissue and is the primary cause of tooth loss in adults. It is mainly caused by subgingival Clostridium nucleatum, Pueraria intermedia, porphyromonas gingivalis and other anaerobic bacteria.At present, there are few reports on the use of natural plants to prevent and cure periodontitis safely. Asarum is a traditional Chinese herbal medicine in China, which has a wide range of pharmacological effects, such as analgesia, cough, anti-inflammation, anti-allergic, anti-oxidation and so on.In this study, the effects of different ratio of liquid to material, distillation time, particle diameter and soaking time on the extraction rate of volatile oil were investigated by single factor experiment.The results showed that the effect on the extraction rate of volatile oil was the particle diameter of the distillation time.The optimal horizontal combination is liquid / material ratio 17, particle diameter 2.5D95 鈮,
