舌鱗狀細(xì)胞癌組織microRNA-29b表達(dá)生物學(xué)意義研究

【摘要】:目的:探討微小RNA-29b(microRNA-29b,miR-29b)在舌鱗狀細(xì)胞癌(tongue squamous cell carcinoma,TSCC)組織中的表達(dá)及其生物學(xué)意義。方法:收集2012-01-02-2013-03-15江西省腫瘤醫(yī)院頭頸外科行手術(shù)切除的45例TSCC及對(duì)應(yīng)癌旁組織(距離切緣2cm),qRT-PCR技術(shù)檢測(cè)miR-29b在TSCC及對(duì)應(yīng)癌旁組織中的表達(dá)情況,并分析其與臨床病理特征的關(guān)系;應(yīng)用miR-29bmimic轉(zhuǎn)染人舌鱗癌Tca-8113細(xì)胞,MTT和流式細(xì)胞技術(shù)檢測(cè)轉(zhuǎn)染后細(xì)胞增殖和凋亡變化,qRT-PCR和蛋白質(zhì)印跡法檢測(cè)miR-29b潛在靶點(diǎn)Bcl-2mRNA及蛋白的表達(dá)變化。結(jié)果:qRT-PCR結(jié)果顯示,miR-29b在TSCC組織中的表達(dá)量為0.213±0.071,顯著低于對(duì)應(yīng)癌旁組織的1.873±0.114,t=2.531,P=0.031;miR-29b低表達(dá)與腫瘤體積增大(≥3cm,χ2=4.022,P=0.033)及高TNM分期(Ⅲ+Ⅳ,χ2=3.398,P=0.036)具有顯著相關(guān)性;MTT結(jié)果顯示,在Tca-8113細(xì)胞中轉(zhuǎn)染miR-29b48h后可顯著降低細(xì)胞增殖能力,P0.05;同時(shí),轉(zhuǎn)染miR-29b72h后細(xì)胞凋亡率由(2.31±0.98)%增加到(11.83±1.36)%,t=2.237,P=0.034。Real-time PCR及蛋白質(zhì)印跡結(jié)果顯示,轉(zhuǎn)染miR-29b后可顯著下調(diào)Tca-8113細(xì)胞內(nèi)Bcl-2mRNA及蛋白表達(dá),P0.05。結(jié)論:TSCC組織中miR-29b表達(dá)下調(diào)并與其臨床病理特征相關(guān),miR-29b可能通過(guò)下調(diào)Bcl-2的表達(dá)來(lái)抑制舌鱗癌Tca-8113細(xì)胞增殖并促進(jìn)其凋亡。
[Abstract]:Objective: to investigate the expression of microRNA-29b squamous miR-29b in tongue squamous cell carcinoma (TSCC) tissue and its biological significance. Methods: 45 cases of TSCC and corresponding adjacent tissues (distance cut) were collected from head and neck surgery of Jiangxi Provincial tumor Hospital from January 2012 to February 15, 2013 to 15, 2013. The expression of miR-29b in TSCC and adjacent tissues was detected by qRT-PCR. The relationship between it and clinicopathological features was analyzed. Cell proliferation and apoptosis were detected by miR-29bmimic transfection of human tongue squamous cell carcinoma Tca-8113 cell line and flow cytometry. MiR-29b potential target Bcl-2mRNA and protein expression were detected by qRT-PCR and Western blotting. The expression level in the tissue was 0.213 鹵0.071, which was significantly lower than that in the corresponding adjacent tissues (1.873 鹵0.114).
