降鈣素上調(diào)TGF-β表達促牙周膜細(xì)胞增殖及膠原合成的研究

【摘要】:目的:研究降鈣素(CT)對人牙周膜細(xì)胞(PDLCs)的增殖、膠原合成的影響及調(diào)控途徑。方法:采用含CT開放閱讀框序列的重組腺病毒Ad.CT轉(zhuǎn)染PDLCs,并以轉(zhuǎn)染空病毒Ad.Null的PDLCs作為對照,分別用流式細(xì)胞術(shù)檢測細(xì)胞周期、定量PCR和Western blot檢測CT、膠原(Ⅰ、Ⅲ、Ⅳ型)、TGF-β的mRNA及蛋白的表達;Ad.CT轉(zhuǎn)染PDLCs同時給予TGF-β抑制劑LY2109761干預(yù),分別檢測細(xì)胞周期及其膠原的表達水平。結(jié)果:轉(zhuǎn)染Ad.CT后PDLCs中CT表達水平較對照組顯著升高(P0.05),G2/S/M期細(xì)胞比例顯著高于對照組(P0.05),Ⅰ、Ⅲ、Ⅳ型膠原蛋白以及TGF-β的表達顯著高于對照組(P0.05)。LY2109761能顯著逆轉(zhuǎn)Ad.CT對于PDLCs增殖及膠原合成的促進作用。結(jié)論:CT可能通過TGF-β途徑促進PDLC細(xì)胞的增殖及膠原的合成。
[Abstract]:Aim: to study the effect of calcitonin on the proliferation and collagen synthesis of human periodontal ligament cells (PDLCs). Methods: PDLCs were transfected with recombinant adenovirus Ad.CT containing CT open reading frame sequence and PDLCs transfected with empty virus Ad.Null as control. The cell cycle was detected by flow cytometry, and CTLs, mRNA and protein expression of TGF- 尾 were detected by quantitative PCR and Western blot. PDLCs was transfected with TGF- 尾 inhibitor LY2109761. Results: after transfection of Ad.CT, the expression of CT in PDLCs was significantly higher than that in control group.
