【摘要】：目的:觀察不同類型慢性根尖周病組織中肥大細(xì)胞(mast cells,MCs)的分布情況,并分析白細(xì)胞介素1β(interleukin-1β,IL-1β)和白細(xì)胞介素17(interleukin-17,IL-17)在MCs上的表達(dá)情況,探討MCs IL-1β和IL-17在根尖周病發(fā)病機(jī)制中的作用。方法:本實(shí)驗(yàn)共收集102例樣本,分為3組:(1)根尖肉芽腫組32例;(2)根尖囊腫組35例;(3)正常對(duì)照組35例。組織標(biāo)本經(jīng)4%中性甲醛緩沖液固定48 h以上;石蠟包埋,組織連續(xù)切片,HE染色,在光學(xué)顯微鏡下觀察其組織學(xué)改變;采用免疫熒光雙染色法,在熒光顯微鏡下觀察各組標(biāo)本組織表達(dá)IL-1β和IL-17的MCs數(shù)目。結(jié)果:兩組慢性根尖周病的炎癥反應(yīng)程度明顯高于正常對(duì)照組(P0.01);根尖囊腫組與根尖肉芽腫組之間炎癥反應(yīng)程度無(wú)顯著差異(P0.05)。與正常對(duì)照組相比,兩組慢性根尖周病組織中IL-1β和IL-17陽(yáng)性MCs數(shù)目均顯著增多(P0.01);根尖囊腫組與根尖肉芽腫組IL-1β和IL-17陽(yáng)性MCs密度無(wú)顯著性差異(P0.05)。經(jīng)Pearson相關(guān)分析,3組標(biāo)本組織中IL-1β和IL-17陽(yáng)性MCs密度與組織炎癥反應(yīng)程度存在直線的正相關(guān)關(guān)系(P0.01)。結(jié)論:慢性根尖周病組織中MCs數(shù)目明顯增多,同時(shí)IL-1β和IL-17陽(yáng)性的MCs密度顯著增高。IL-1β和IL-17陽(yáng)性MCs密度與根尖周病的炎癥程度趨勢(shì)相一致。這提示IL-1β和IL-17陽(yáng)性MCs可能在慢性根尖周病的致病機(jī)制中發(fā)揮重要作用。
[Abstract]:Objective: to observe the distribution of mast cell mast cells in different types of chronic periapical disease, and to analyze the expression of interleukin-1 尾 interleukin-1 尾 interleukin-1 尾 (IL-1 尾) and interleukin-17interleukin-17IL-17 (IL-17) on MCs. To investigate the role of MCs IL-1 尾 and IL-17 in the pathogenesis of periapical disease. The specimens were fixed with 4% neutral formaldehyde buffer solution for more than 48 hours, embedded in paraffin wax, stained with HE in tissue sections. The histological changes were observed under an optical microscope. The number of MCs expressing IL-1 尾 and IL-17 was observed under fluorescence microscope. Results: the inflammatory reaction degree of chronic periapical disease in the two groups was significantly higher than that in the control group (P 0.01), and the inflammatory reaction between the apical cyst group and the apical granuloma group was significantly higher than that in the control group. There was no significant difference in the degree of adaptation (P 0.05). Compared with the normal control group, there was no significant difference between the two groups. The number of IL-1 尾 and IL-17 positive MCs in the two groups of chronic periapical disease was significantly increased (P 0.01), the density of IL-1 尾 and IL-17 positive MCs was not significantly different between the apical cyst group and the apical granuloma group. The Pearson correlation analysis showed that IL-1 尾 and IL-17 were positive in the three groups. There was a linear positive correlation between the density of MCs and the degree of inflammatory response. Conclusion: the number of MCs in chronic periapical disease tissues increased significantly. At the same time, the MCs density of IL-1 尾 and IL-17 positive MCs increased significantly. IL-1 尾 and IL-17 positive MCs density were consistent with the inflammatory degree of periapical disease, which suggested that IL-1 尾 and IL-17 positive MCs might play an important role in the pathogenesis of chronic periapical disease.
