【摘要】：目的炎癥通過其對(duì)腫瘤微環(huán)境的影響在癌癥的發(fā)展過程中起著重要作用。然而炎癥與腫瘤之間的關(guān)系仍然需要進(jìn)一步闡明。本研究主要觀察IL-21R在舌鱗狀細(xì)胞癌的表達(dá)和分布情況。方法使用逆轉(zhuǎn)錄酶聚合酶鏈反應(yīng)(RT-PCR)、Western blot免疫印跡方法檢測(cè)來自54個(gè)舌鱗狀細(xì)胞癌患者手術(shù)標(biāo)本IL-21R的基因和蛋白表達(dá),并且觀察IL-21R表達(dá)與臨床病理特征的關(guān)系。同時(shí),進(jìn)一步通過免疫組織化學(xué)和免疫熒光技術(shù)觀察IL-21R表達(dá)與分布規(guī)律。結(jié)果 RT-PCR和Western印跡分析的結(jié)果表明,與癌旁組織相比,舌鱗狀細(xì)胞癌標(biāo)本中IL-21R基因和蛋白表達(dá)顯著降低(P0.05)。IL-21R低表達(dá)與性別、年齡、分化程度、腫瘤大小均無密切關(guān)系。這種表達(dá)特點(diǎn)主要與臨床分期呈正相關(guān)。免疫組織化學(xué)結(jié)果顯示,IL-21R主要表達(dá)于舌癌旁組織鱗狀細(xì)胞和上皮細(xì)胞,而鱗狀細(xì)胞癌組織中表達(dá)較弱或散在的表達(dá)。IL-21R與IL-10的雙染熒光結(jié)果顯示,IL-21R陽性的細(xì)胞中可見IL-10的陽性染色。結(jié)論這些數(shù)據(jù)表明IL-21R在舌癌腫瘤發(fā)生與侵襲性過程中可能起著重要作用。
[Abstract]:The purpose of inflammation through its impact on the tumor microenvironment in cancer development plays an important role. However, the relationship between inflammation and cancer remains to be elucidated. This study is designed to investigate the expression and distribution of IL-21R in tongue squamous cell carcinoma. Methods using reverse transcriptase polymerase chain reaction (RT-PCR), gene and protein expression detection of Western blot by Western blot method from 54 surgical specimens of patients with tongue squamous cell carcinoma cell line IL-21R, and observe the relationship between IL-21R expression and clinicopathological features. At the same time, further through the observation of IL-21R chemical and immunofluorescence techniques in immune tissues and distribution. The results of RT-PCR and Western blot analysis showed that compared with the adjacent tissues. Expression of IL-21R in specimens of squamous cell carcinoma of the tongue gene and protein significantly decreased (P0.05) low expression of.IL-21R and gender, age, differentiation degree, tumor large Small had no close relationship. The expression characteristics and clinical stage were positively correlated. Immunohistochemical results showed that IL-21R was mainly expressed in tissues of squamous cell carcinoma of tongue and epithelial cells, squamous cell carcinoma and expression of.IL-21R and IL-10 is weak or scattered in the fluorescent double staining showed that positive staining of IL-10 showed IL-21R positive the cells. Conclusion these data suggest that IL-21R may play an important role in tumorigenesis and invasive squamous cell carcinoma of the tongue in the process.
