新疆墨玉地區(qū)維吾爾族成人中、重度慢性牙周炎與雌激素受體基因多態(tài)性的相關(guān)性研究

【摘要】:目的由于地域、民族等不同因素影響,雌激素受體基因多態(tài)性與牙周疾病的關(guān)系尚未明確。文中探討雌激素受體α、β多態(tài)性與新疆墨玉地區(qū)維吾爾族成人中、重度慢性牙周炎易感性的關(guān)系。方法選取2013年3-4月在墨玉地區(qū)流行病學(xué)調(diào)查并收集的97例中、重度慢性牙周炎和102例健康對照者的口腔頰黏膜細(xì)胞樣本。所有研究對象均為維吾爾族,年齡18~70歲。牙周炎組納入標(biāo)準(zhǔn):口內(nèi)至少有10顆牙可以進(jìn)行牙周檢查,其中包括4顆磨牙(不含第三磨牙),所有牙周炎組個體均無系統(tǒng)性疾病及已知可影響牙周狀況的疾病。對照組納入標(biāo)準(zhǔn):無系統(tǒng)性疾病,口腔衛(wèi)生情況良好,牙齦無異常,無附著喪失,牙齒無松動,缺失牙不超過3顆。2組研究對象均排除懷孕女性。提取全基因組DNA,用聚合酶鏈反應(yīng)-限制性片段長度多態(tài)性技術(shù)檢測雌激素受體α、β基因型分布。結(jié)果牙周炎組與對照組比較,雌激素受體αXbaⅠ基因型表達(dá)差異無統(tǒng)計學(xué)意義(P=0.180 9),但等位基因頻率差異有統(tǒng)計學(xué)意義(P=0.036 6),PvuⅡ基因型表達(dá)和等位基因頻率差異均有統(tǒng)計學(xué)意義(P=0.021 5、P=0.014 6);雌激素受體βRsaⅠ基因型表達(dá)和等位基因頻率差異均無統(tǒng)計學(xué)意義(P=0.180 9、P=0.094 9),AluⅠ基因型表達(dá)差異無統(tǒng)計學(xué)意義(P=0.059 4),但等位基因頻率差異有統(tǒng)計學(xué)意義(P=0.048 3)。單因素Logistic回歸模式分析顯示pp基因型可能為該地區(qū)患牙周炎的危險因素之一(P=0.051),Aa基因型為該地區(qū)患牙周炎的保護(hù)因素(P=0.032)。2組女性研究對象僅在雌激素受體αPvuⅡ等位基因頻率差異有統(tǒng)計學(xué)意義(P=0.022 9),而男性在雌激素受體α、β中差異未見統(tǒng)計學(xué)意義(P0.05)。結(jié)論新疆墨玉地區(qū)維吾爾族成人中、重度慢性牙周炎易感性與雌激素受體α、β基因型分布相關(guān)。雌激素受體αPvuⅡ中pp基因型可能是維吾爾族慢性牙周炎的易感因子。
[Abstract]:Objective to explore the relationship between estrogen receptor gene polymorphism and periodontal disease due to different factors, such as region, nationality, etc. In this paper, the relationship between estrogen receptor 偽, 尾 polymorphism and Uygur adults in Moyu area, Xinjiang, was discussed. Methods 97 cases of severe chronic periodontitis were investigated and collected in Moyu area in 2013. Oral and buccal mucosal cell samples from patients with severe chronic periodontitis and 102 healthy controls. All subjects were Uygur, aged 1870. Periodontitis group was included in the criteria: at least 10 teeth in the mouth could be examined for periodontal examination. These included 4 molars (excluding the third molar, no systemic disease in all periodontitis individuals and known diseases affecting periodontal status. Control group included: no systemic disease, good oral hygiene, no gingival abnormality, No loss of attachment, no loosening of teeth, The whole genome DNA was extracted and the distribution of estrogen receptor 偽, 尾 genotypes was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique.
