【摘要】：目的：采用紫杉醇類聯(lián)合卡鉑經(jīng)不同途徑灌注治療兔VX-2舌癌，比較療效的差異。 方法：建立兔VX-2舌癌動(dòng)物模型，隨機(jī)分為6組，每組4只，分別為：A1（紫杉醇脂質(zhì)體聯(lián)合卡鉑舌動(dòng)脈灌注給藥組），A2（紫杉醇聯(lián)合卡鉑舌動(dòng)脈灌注給藥組），A（葡萄糖舌動(dòng)脈灌注給藥組），V1（紫杉醇脂質(zhì)體聯(lián)合卡鉑全身靜脈給藥組），，V2（紫杉醇聯(lián)合卡鉑全身靜脈給藥組），V（葡萄糖全身靜脈給藥組）�；熃o藥時(shí)間1小時(shí)�；�1周后通過測量舌癌原發(fā)灶體積、流式細(xì)胞術(shù)、免疫組化的方法評(píng)價(jià)腫瘤抑瘤率、細(xì)胞凋亡情況、p53蛋白表達(dá)情況，各組數(shù)據(jù)用SPSS20.0統(tǒng)計(jì)軟件進(jìn)行分析，比較紫杉醇類聯(lián)合卡鉑經(jīng)不同給藥途徑治療兔VX-2舌癌的療效。 結(jié)果：①化療后各組有效率分別為：A1組4/4、A2組1/4，V1、V2、A、V組0/4。A1組與V1組、V2組、A組、V組組間比較具有顯著性差異（P0.05）；余組間比較無顯著性差異（P0.05）。②經(jīng)FCM檢測腫瘤細(xì)胞凋亡率（%）：A1組：84.80±3.35、A2組：62.70±7.92、A組：8.63±0.38、V1組：52.05±3.96、V2組：29.45±0.15、V組：8.61±0.28，A1組明顯高于其它各組，各組間比較具有顯著性差異（P0.05）。③免疫組化測得P53蛋白在腫瘤組織的表達(dá)：在A1、A2、V1、V2、A、V的陽性表達(dá)率分別為8.33%、33.33%、58.33%、66.67%、83.33%、83.33%。A1組分別與V1組、V2組、A組、V組組間比較具有顯著性差異（P0.05）；A2組分別與A組、V組組間比較具有顯著性差異（P0.05）；余各組間比較無顯著性差異（P0.05）。 結(jié)論：①紫杉醇脂質(zhì)體聯(lián)合卡鉑化療較紫杉醇聯(lián)合卡鉑治療兔VX-2舌癌近期療效顯著；②經(jīng)舌動(dòng)脈灌注化療較全身靜脈化療治療兔VX-2舌癌近期療效顯著。
[Abstract]:Objective: To compare the therapeutic effect of paclitaxel and carboplatin through different ways in the treatment of VX-2 tongue cancer in rabbits.
Methods: rabbit tongue VX-2 animal model, randomly divided into 6 groups, 4 rats in each group, respectively: A1 (lingual arterial infusion of paclitaxel liposome combined with Carboplatin Injection group (A2), lingual artery perfusion with paclitaxel and carboplatin administration group (A), glucose tongue arterial infusion group), V1 (paclitaxel liposome combined with carboplatin in the intravenous group), V2 (paclitaxel combined with carboplatin in the intravenous group), V (glucose intravenous group). Chemotherapy was administered for 1 hours. After 1 weeks of chemotherapy by measuring the volume of primary squamous cell carcinoma of the tongue, flow cytometry, method of immunohistochemical evaluation of tumor inhibition rate, cell apoptosis, the expression of p53 protein were analyzed by SPSS20.0 statistical software, all the data were compared with paclitaxel and carboplatin in the curative effect of different routes of Administration for the treatment of rabbit VX-2 carcinoma of tongue.
