上頜竇區(qū)種植術(shù)骨高度不足的臨床處理及基于3D打印的微納復(fù)合結(jié)構(gòu)鈦種植體對成骨的影響

【摘要】:目的:研究鈦表面微納復(fù)合結(jié)構(gòu)對大鼠骨髓間充質(zhì)干細(xì)胞黏附、增殖以及成骨分化的影響。方法:采用水熱法利用鈦酸四丁酯為鈦源,在酸蝕過的3D打印鈦片上生成二氧化鈦納米棒結(jié)構(gòu)。其表面微納復(fù)合拓?fù)浣Y(jié)構(gòu)采用掃描電鏡(SEM)進(jìn)行檢測。通過倒置熒光顯微鏡觀察、CCK-8輔助檢測不同表面細(xì)胞黏附、增殖情況。采用Realtime PCR檢測各組細(xì)胞成骨分化相關(guān)基因RUNX2、ColⅠ、OTC的表達(dá)水平。結(jié)果:SEM可見微納復(fù)合結(jié)構(gòu)鈦片表面納米級棒狀結(jié)構(gòu),酸蝕過的3D打印鈦片表面具有微米尺度的粗糙度,金鋼砂紙打磨普通鈦片表面較為平整光滑。各組孵育大鼠BMSCs24h后檢測示微納復(fù)合結(jié)構(gòu)對細(xì)胞伸展更加有利,而黏附的細(xì)胞數(shù)低于普通鈦片組,cck-8結(jié)果與之相符;72h后檢測示微納復(fù)合結(jié)構(gòu)不僅促進(jìn)細(xì)胞增殖遠(yuǎn)高于普通鈦片組,細(xì)胞平鋪展開面積也是其兩倍,而3D鈦片酸蝕組各項(xiàng)均處于中間值。第七天檢測成骨基因表達(dá)水平示,3D鈦片酸蝕組和微納復(fù)合結(jié)構(gòu)組三種基因的表達(dá)都遠(yuǎn)多于普通鈦片組,差異具有顯著性,同時微納復(fù)合結(jié)構(gòu)組三種基因表達(dá)均高于3D鈦片酸蝕組。結(jié)論:微納復(fù)合結(jié)構(gòu)有利于細(xì)胞增殖以及細(xì)胞伸展,同時更利于細(xì)胞向成骨方向分化。推測其可能原因在于,基于3D打印所制備的微米表面具有更易為細(xì)胞所識別的粗糙度,同時,微納復(fù)合結(jié)構(gòu)為細(xì)胞提供了生理三維仿生環(huán)境,有利于細(xì)胞鋪展以促進(jìn)其更好地增殖與分化。
[Abstract]:Objective: to study the effect of titanium surface microcomposite structure on the adhesion, proliferation and osteogenic differentiation of rat bone marrow mesenchymal stem cells. Methods: tetrabutyl titanate was used as titanium source by hydrothermal method. Titanium dioxide nanorods were formed on the acid etched 3D printed titanium wafer.
