AKT-Foxo3a信號(hào)通路調(diào)控小鼠MMP-20基因表達(dá)的研究

【摘要】:目的:本研究旨在探討AKT-Foxo3a信號(hào)通路在基質(zhì)金屬蛋白酶-20(matrix metalloproteinase-20,MMP-20)基因表達(dá)中的作用。方法:分別通過(guò)免疫組化技術(shù)、實(shí)時(shí)定量RT-PCR技術(shù)以及雙熒光素酶報(bào)告基因檢測(cè)系統(tǒng)分析AKT-Foxo3a信號(hào)通路對(duì)MMP-20啟動(dòng)子轉(zhuǎn)錄活性的影響。結(jié)果:免疫組化染色結(jié)果顯示,AKT及磷酸化Foxo3a(P-Foxo3a)均在分泌期成釉細(xì)胞核中呈陽(yáng)性表達(dá);RT-PCR檢測(cè)結(jié)果顯示,AKT阻斷劑可顯著抑制Foxo3a誘導(dǎo)的MMP-20基因的表達(dá)(P㩳0.05);雙熒光素酶分析結(jié)果顯示,AKT阻斷劑亦可顯著抑制Foxo3a調(diào)控MMP-20基因啟動(dòng)子的轉(zhuǎn)錄活性(P㩳0.05)。結(jié)論:轉(zhuǎn)錄因子Foxo3a可能通過(guò)AKT-Foxo3a信號(hào)通路調(diào)控MMP-20基因的表達(dá),從而參與釉質(zhì)發(fā)育過(guò)程。
[Abstract]:Objective: to investigate the role of AKT-Foxo3a signaling pathway in the expression of matrix metalloproteinase-20 (MMP-20) gene. Real-time quantitative RT-PCR technique and double luciferase reporter gene detection system were used to analyze the effect of AKT-Foxo3a signaling pathway on the transcriptional activity of MMP-20 promoter. Results: the results of immunohistochemical staining showed that the ameloblast in secretory phase was ameloblast with phosphorylated Foxo3a and phosphorylated Foxo3a. The results of reverse transcription-polymerase chain reaction (RT-PCR) showed that Foxo3a antagonist could significantly inhibit the expression of MMP-20 gene induced by Foxo3a. The results of double luciferase analysis showed that AK antagonist could significantly inhibit the transcriptional activity of MMP-20 promoter regulated by Foxo3a. Conclusion: the transcription factor Foxo3a may regulate the expression of MMP-20 gene through AKT-Foxo3a signaling pathway and thus participate in the development of enamel.
