【摘要】：目的:探討細胞周期調(diào)控蛋白激酶1(cyclin-dependent kinases 1,CDK1)在口腔鱗癌患者中的臨床意義及相關(guān)miR-192在口腔鱗癌中表達。方法:采用qPCR法檢測8例口腔鱗癌組織中miR-192的表達,并分析其在口腔鱗癌中表達的趨勢;同時用SP免疫組織化學(xué)測定77例口腔鱗癌標本CDK1蛋白的表達,并結(jié)合患者的術(shù)后復(fù)發(fā)、轉(zhuǎn)移及5年生存率情況進行相關(guān)分析。實驗結(jié)果:對比正常組織組,miR-192及pri-miR-192在口腔鱗癌中表達增加,miR-192更明顯(P=0.0005160.01)。CDK1蛋白陽性表達與口腔鱗癌患者術(shù)后復(fù)發(fā)及淋巴轉(zhuǎn)移關(guān)系密切,并有統(tǒng)計學(xué)意義(P0.05);同時,患者CDK1蛋白陽性組的5年生存率顯著低于陰性組,兩組之間有統(tǒng)計學(xué)意義(P0.05),陽性組患者預(yù)后更差。結(jié)論:評估口腔鱗癌患者CDK1蛋白的表達或許可作為預(yù)測口腔鱗癌患者預(yù)后的指標之一。miR-192在口腔鱗癌中表達增加,與正常組織相比有顯著性差異;可進一步結(jié)合miR-192與CDK1共同預(yù)測口腔鱗癌的預(yù)后。
[Abstract]:Objective: to investigate the clinical significance of cyclin dependent kinases 1 (CDK1) in oral squamous cell carcinoma (OSCC) and the expression of miR-192 in oral squamous cell carcinoma (OSCC). Methods: qPCR method was used to detect the expression of miR-192 in 8 cases of oral squamous cell carcinoma (OSCC). At the same time, the expression of CDK1 protein in 77 cases of oral squamous cell carcinoma was detected by SP immunohistochemical method, and combined with the postoperative recurrence of the patients, the expression of CDK1 protein in oral squamous cell carcinoma (OSCC) was analyzed. Results: compared with the normal tissue group, the expression of miR-192 and pri-miR-192 in oral squamous cell carcinoma (OSCC) was significantly higher than that in normal tissue group (P < 0. 000516. 01). CDK1 protein expression was closely related to postoperative recurrence and lymphatic metastasis in patients with oral squamous cell carcinoma (OSCC). The 5-year survival rate in CDK1 positive group was significantly lower than that in negative group. Conclusion: the expression of CDK1 protein in patients with oral squamous cell carcinoma may be one of the predictors of prognosis of oral squamous cell carcinoma, and the expression of miR-192 is increased in oral squamous cell carcinoma. The prognosis of oral squamous cell carcinoma can be predicted by combining miR-192 and CDK1.
