ERK1/2及CRF系統(tǒng)在可卡因成癮過程中的作用及其機(jī)制研究

研究背景可卡因是一種具有興奮中樞神經(jīng)系統(tǒng)作用的局部麻醉藥。因?yàn)樗梢砸鹁衽d奮、產(chǎn)生快感,所以成為吸毒者濫用的毒品之一。近年來,隨著實(shí)驗(yàn)手段的飛躍發(fā)展,科研工作者從神經(jīng)心理學(xué)、神經(jīng)生理學(xué)、發(fā)育生物學(xué)、生物化學(xué)和分子生物學(xué)等各個方面對可卡因的神經(jīng)毒性作用機(jī)制進(jìn)行了廣泛深入的研究?煽ㄒ?yàn)E用可導(dǎo)致中樞的鎬賞和動機(jī)相關(guān)的神經(jīng)環(huán)路發(fā)生可塑性變化,提示可卡因成癮是一種生理功能和精神紊亂疾病。目前,有關(guān)可卡因神經(jīng)毒性作用的腦區(qū)的研究主要集中在多巴胺能神經(jīng)通路。其中,中腦-邊緣葉通路(由中腦腹后被蓋區(qū)投射到腹側(cè)紋狀體和邊緣系統(tǒng)),中腦-大腦皮質(zhì)通路及黑質(zhì)-紋狀體通路為研究的主要靶點(diǎn)。這三條通路中,紋狀體成為重要的交通關(guān)聯(lián)要道。目前公認(rèn)的可卡因毒性機(jī)理多巴胺學(xué)說認(rèn)為:可卡因及其代謝產(chǎn)物主要通過阻斷多巴胺轉(zhuǎn)運(yùn)體的活動,引起突觸間隙多巴胺聚集增多,進(jìn)而改變突觸前后多巴胺受體活動發(fā)揮神經(jīng)毒性作用的。但有關(guān)濫用可卡因如何引起神經(jīng)環(huán)路發(fā)生適應(yīng)性變化的分子調(diào)控機(jī)制的研究還十分貧乏。近年,胞外信號調(diào)節(jié)蛋白激酶(Extracellular signal-regulated protein kinase,ERK),尤...
