ERK1/2及CRF系統(tǒng)在可卡因成癮過程中的作用及其機制研究

研究背景可卡因是一種具有興奮中樞神經系統(tǒng)作用的局部麻醉藥。因為它可以引起精神興奮、產生快感,所以成為吸毒者濫用的毒品之一。近年來,隨著實驗手段的飛躍發(fā)展,科研工作者從神經心理學、神經生理學、發(fā)育生物學、生物化學和分子生物學等各個方面對可卡因的神經毒性作用機制進行了廣泛深入的研究?煽ㄒ驗E用可導致中樞的鎬賞和動機相關的神經環(huán)路發(fā)生可塑性變化,提示可卡因成癮是一種生理功能和精神紊亂疾病。目前,有關可卡因神經毒性作用的腦區(qū)的研究主要集中在多巴胺能神經通路。其中,中腦-邊緣葉通路(由中腦腹后被蓋區(qū)投射到腹側紋狀體和邊緣系統(tǒng)),中腦-大腦皮質通路及黑質-紋狀體通路為研究的主要靶點。這三條通路中,紋狀體成為重要的交通關聯(lián)要道。目前公認的可卡因毒性機理多巴胺學說認為:可卡因及其代謝產物主要通過阻斷多巴胺轉運體的活動,引起突觸間隙多巴胺聚集增多,進而改變突觸前后多巴胺受體活動發(fā)揮神經毒性作用的。但有關濫用可卡因如何引起神經環(huán)路發(fā)生適應性變化的分子調控機制的研究還十分貧乏。近年,胞外信號調節(jié)蛋白激酶(Extracellular signal-regulated protein kinase,ERK),尤...
