大鼠內(nèi)側(cè)隔核注射淀粉樣β蛋白影響海馬theta節(jié)律和長時(shí)程增強(qiáng)的在體電生理研究

阿爾茨海默。╝lzheimer’s diseases, AD)是一種在老年人中常見的癡呆病,臨床表現(xiàn)是進(jìn)行性發(fā)展的認(rèn)知功能減退,以細(xì)胞外沉積的老年斑、細(xì)胞內(nèi)神經(jīng)原纖維纏結(jié)和神經(jīng)元喪失為病理特征。在AD,大腦的皮層和海馬都有大量的老年斑沉積,老年斑的主要成分是淀粉樣β蛋白(amyloidβ-protein, Aβ),其來源于淀粉樣前體蛋白(amyloid precursor protein, APP)的酶解。自然產(chǎn)生的Aβ包含40或42個(gè)氨基酸,具有廣泛的神經(jīng)毒性。實(shí)驗(yàn)發(fā)現(xiàn),人工合成的Aβ25-35,甚至氨基酸數(shù)量更少的Aβ31-35也同樣具有神經(jīng)毒性。Aβ的產(chǎn)生和集聚被普遍認(rèn)為是造成AD的主要病因。進(jìn)一步用轉(zhuǎn)基因小鼠的實(shí)驗(yàn)表明,除了皮層和海馬,隔核也有Aβ的廣泛沉積。同時(shí),在AD病人,包括隔核在內(nèi)的基底膽堿能系統(tǒng)神經(jīng)元也有顯著的喪失。因此,基底前腦膽堿能系統(tǒng)的受損,被認(rèn)為在很大程度上與AD病人所出現(xiàn)的認(rèn)知缺陷相關(guān),此即AD病因的膽堿能假說。海馬theta節(jié)律是海馬局部場電位(local field potential, LFP)中一個(gè)頻率在3-12 Hz的近似于正弦波的成分。據(jù)認(rèn)為,...
