Alzheimer病模型小鼠海馬代謝物早期變化的9.4T磁共振頻譜研究

目的:通過9.4T磁共振頻譜離體檢測阿爾茨海默。ˋlzheimer’s disease,AD)模型小鼠疾病過程中的代謝物變化,以找出MRS早期診斷此病的生物學(xué)標(biāo)識。方法: 36只昆明小鼠隨機(jī)分成5個實(shí)驗(yàn)組和1個對照組,每組6只,對于實(shí)驗(yàn)組各組小鼠每天進(jìn)行皮下注射D-半乳糖120mg/(kg.d)和亞硝酸鈉90mg/(kg.d),各組分別連續(xù)注射30天、40天、50天、60天及70天,對照組小鼠每天注射相同劑量的生理鹽水70天,藥物注射結(jié)束后行水迷宮測試(MWM)并犧牲小鼠取其兩側(cè)海馬。右側(cè)海馬經(jīng)高氯酸萃取等處理后凍干成粉末,利用9.4T磁共振進(jìn)行頻譜測試觀察代謝物變化;左側(cè)海馬用來做病理組織學(xué)檢測。對于代謝物氮-乙酰天門冬氨酸(N-acetylaspartate, NAA),谷氨酸(glutamate, Glu),肌醇(myo-inositol, mI)和膽堿(choine, Cho)進(jìn)行分析。結(jié)果:水迷宮檢查結(jié)果顯示實(shí)驗(yàn)組70天時空間學(xué)習(xí)及記憶能力較對照組明顯降低(P<0.05)。電鏡下觀察病理染色圖片可見60天及70天之小鼠海馬區(qū)神經(jīng)元細(xì)胞明顯減少,變性,神經(jīng)原纖維纏結(jié)及老...
