遠(yuǎn)志總皂苷對(duì)阿爾茨海默病大鼠突觸可塑性的影響

【摘要】:目的: 1觀察遠(yuǎn)志總皂苷對(duì)AD大鼠在體海馬CA_1區(qū)神經(jīng)電生理指標(biāo)長(zhǎng)時(shí)程增強(qiáng)的影響。 2觀察遠(yuǎn)志總皂苷對(duì)AD大鼠海馬CA_1區(qū)N-甲基-D-天冬氨酸受體2A亞基表達(dá)的影響。方法: 雄性Wistar大鼠隨機(jī)分為對(duì)照組、模型組、高劑量遠(yuǎn)志總皂苷治療組(37.5 mg/mL)和低劑量遠(yuǎn)志總皂苷治療組(12.5 mg/mL),每組8只,造模方法為腹腔注射D-半乳糖引起大鼠早衰,再用鵝膏蕈氨酸損毀Meynert核誘導(dǎo)大鼠發(fā)生AD的病理變化,,測(cè)定并比較各組大鼠神經(jīng)電生理高頻刺激(HFS)前后在體海馬CA_1區(qū)場(chǎng)興奮性突觸后電位(fEPSP)幅度的變化;測(cè)定海馬CA_1免疫組織化學(xué)NR2AIOD值間接測(cè)定其表達(dá)。 結(jié)果: 1、遠(yuǎn)志總皂苷對(duì)AD大鼠在體海馬CA_1區(qū)神經(jīng)電生理指標(biāo)長(zhǎng)時(shí)程增強(qiáng)的影響:對(duì)照組fEPSP的幅度值在HFS 1分鐘、30分鐘和60分鐘分別達(dá)到基線水平的203.17±7.47%、178.15±8.11%和164.17±7.03%水平,而模型組fEPSP幅度較對(duì)照組顯著降低(p0.05),僅分別達(dá)到基線水平的173.63±10.81%、127.12±7.38%和102.88±2.36%。低劑量遠(yuǎn)志總皂苷組fEPSP的幅度百分比值較模型組升高(p0.05),分別達(dá)到基線水平的177.67±14.04%、131.83±4.96%和121.17±4.79%,高劑量遠(yuǎn)志總皂苷組fEPSP的幅度百分比值較模型組顯著升高(p0.05),分別達(dá)到基線水平的192.00±2.45%、168.00±2.45%和141.75±9.25%,且高劑量遠(yuǎn)志總皂苷組fEPSP的幅度百分比值較低劑量組升高(p0.05)。 2、遠(yuǎn)志總皂苷對(duì)AD大鼠海馬CA_1區(qū)突觸結(jié)構(gòu)N-甲基-D-天冬氨酸受體2A亞基表達(dá)的影響:對(duì)照組、AD組、低、高劑量組的NR2A積分光密度值(IOD值)分別為30.12±3.45、11.74±1.69、20.78±2.66、25.86±2.98。與對(duì)照組比較,AD模型組IOD值明顯降低(p0.05),間接反映了其表達(dá)水平降低。與模型組比較,遠(yuǎn)志總皂苷高、低劑量組的IOD值增高,間接反映其表達(dá)水平明顯增高(p0.05, p0.05),并且高劑量組NR2A的表達(dá)較低劑量組顯著(p0.05)。 結(jié)論: 1、遠(yuǎn)志總皂苷對(duì)AD大鼠海馬CA_1區(qū)神經(jīng)電生理指標(biāo)LTP具有易化作用。 2、遠(yuǎn)志總皂苷可上調(diào)AD大鼠海馬CA_1區(qū)突觸結(jié)構(gòu)N-甲基-D-天冬氨酸受體2A亞基表達(dá)。 3、遠(yuǎn)志總皂苷對(duì)上述兩作用呈現(xiàn)劑量依賴性。
[Abstract]:Aim: 1 to observe the effect of total saponins of Polygaloside on long-term potentiation of neuroelectrophysiological indexes in CA _ 1 region of hippocampus in AD rats. 2 to observe the effect of total saponins of Polygaloside on the expression of N-methyl-D-aspartic acid receptor 2A subunit in hippocampal CA_ 1 region of AD rats. Methods: male Wistar rats were randomly divided into control group, model group, high dose Polygaloside treatment group (37.5 mg/mL) and low dose Polygala saponins treatment group (12.5 mg/mL), with 8 rats in each group. The model was induced by intraperitoneal injection of D-galactose to induce premature senility in rats, and the pathological changes of AD were induced by the destruction of Meynert nucleus by gossypol fungofunc acid. The changes of (fEPSP) amplitude of excitatory postsynaptic potential in hippocampal CA_1 region in vivo before and after high frequency neuroelectrophysiological stimulation of (HFS) were measured and compared. The expression of CA_1 in hippocampus was measured indirectly by NR2AIOD. Results: 1. The effect of total saponins of Polygala tenuifolia on the long-term potentiation of neuroelectrophysiological indexes in hippocampal CA _ 1 region of AD rats: the amplitude of fEPSP in the control group reached 203.17 鹵7.47% of the baseline level at 1 minute, 30 minutes and 60 minutes, respectively. The levels of fEPSP in the model group were significantly lower than those in the control group (p0.05), only 173.63 鹵10.81%, 127.12 鹵7.38% and 102.88 鹵2.36%, respectively. The percentage value of fEPSP in the low-dose total saponin group was higher than that in the model group (p0.05), reaching 177.67 鹵14.04%, 131.83 鹵4.96% and 121.17 鹵4.79% of the baseline level, respectively. The percentage value of fEPSP in the high-dose total saponins group was significantly higher than that in the model group (p0.05), reaching 192.00 鹵2.45% of the baseline level, 192.00 鹵2.45% and 141.75 鹵9.25% of the baseline level, respectively. The amplitude percentage ratio of fEPSP in high dose Polygala tenuifolia group was higher than that in low dose group (p0.05). 2. Effects of total saponins of Polygaloside on the expression of N-methyl-D-aspartate receptor 2A subunit in hippocampal CA_ 1 region of AD rats: control group, AD group, low, low. The NR2A integral optical density (IOD) of high dose group was 30.12 鹵3.45,11.74 鹵1.69, 20.78 鹵2.66, 25.86 鹵2.98, respectively. Compared with the control group, the IOD value of AD model group was significantly lower (p0.05), which indirectly reflected the decrease of IOD expression level. Compared with the model group, the Iod value of total saponins in high dose group and low dose group was significantly higher than that in low dose group (p0.05, p0.05), and the expression of NR2A in high dose group was significantly higher than that in low dose group (p0.05). Conclusion: 1. Polygala tenuifolia saponins can facilitate the neuroelectrophysiological index LTP in hippocampal CA_1 region of AD rats. 2. Polygaloside could up-regulate the expression of N-methyl-D-aspartate receptor 2A subunit in the synaptic structure of hippocampal CA_ 1 region of AD rats. 3. The total saponins of Polygala tenuifolia showed a dose-dependent effect on the above two effects.
