應(yīng)激對SAMP8小鼠認(rèn)知功能及情緒的影響及兩者機(jī)制研究
[Abstract]:Objective to study the effects of chronic unpredictable mild stress (UCMS) on depression, learning and memory ability in SAMP8 mice, and the effect of UCMS on brain-derived neurotrophic factor (BDNF) and its receptor (Trk B), in hippocampus. Effect of apoptosis-related factor B lymphocyte tumor-2 gene (Bcl-2) and Bcl-2-associated X protein (Bax) protein expression. Methods Twenty healthy 7-month-old male SAMP8 mice were randomly divided into two groups: stress group (n = 10) and non-stress group (n = 10). Two groups of mice were subjected to chronic unpredictable stress for 21 days. After the stress was completed, the water maze test and sugar-water preference test were performed. After the behavioral experiment was completed, six mice in the stress group and the non-stress group were randomly selected and the hippocampus was decapitated. The protein expression of BDNF, Trk-B, Bcl-2 and Bax were measured by protein blot (westernblotting). Results (1) after 5 days of training, the latency of finding the platform in the stress group increased significantly (P0.05). After removing the platform on the 6th day, the number of crossing the platform in the stress group was significantly lower than that in the non-stress group (P0.05). 2 the consumption of sugar water in the stress group was significantly lower than that in the non-stress group (P0.05). 3Compared with the non-stress group, the frequency of crossing the platform in the stress group decreased significantly (P0.05). (4) compared with non-stress group, the expression of BDNF and Trk B protein in hippocampus of stress group decreased significantly, and the expression of Bax protein increased significantly, compared with non-stress group, the expression of Bcl-2 and Bax protein in hippocampus of stress group was significantly decreased, and the expression of Bax protein was significantly higher than that of non-stress group. And has statistical significance (P0.05). Conclusion stress induced depression and decreased the ability of learning and memory in SAMP8 mice, the expression of Bcl-2 protein and Bcl-2 protein decreased and the expression of Bax protein increased in BDNF and Trk-B induced by stress.
