NMDA受體NR1亞單位與GABA_A受體在精神分裂癥小鼠海馬齒狀回顆粒細(xì)胞層的表達(dá)

【摘要】:目的:明確NMDA受體NR1亞單位、GABAA受體與精神分裂癥(SP)小鼠海馬齒狀回顆粒細(xì)胞層新生顆粒細(xì)胞的共存模式;闡明NR1、GABAA受體在SP小鼠海馬中的表達(dá)。方法:實(shí)驗(yàn)組小鼠腹腔注射MK-801(每日0.6 mg/kg),對(duì)照組注射等量的生理鹽水,連續(xù)注射14 d后對(duì)兩組動(dòng)物分別進(jìn)行BrdU標(biāo)記,斷頭并進(jìn)行如下處理:(1)免疫熒光染色,觀察海馬DG區(qū)中NR1和GABAA的表達(dá)以及與BrdU標(biāo)記的新生顆粒細(xì)胞的共存模式;(2)利用RT-PCR技術(shù),檢測(cè)海馬GABAA及NR1 mRNA表達(dá)水平的改變。結(jié)果:(1)停藥后實(shí)驗(yàn)組與對(duì)照組比較,海馬神經(jīng)細(xì)胞的增殖率下降了23.1%(P0.05),GABAA、NR1兩種神經(jīng)細(xì)胞增殖數(shù)無(wú)差異性改變(P0.05);(2)實(shí)驗(yàn)組小鼠海馬GABAAmRNA的表達(dá)量較對(duì)照組顯著下降(P0.05),而NR1 mRNA的表達(dá)量較對(duì)照組明顯增高(P0.05)。結(jié)論:(1)小鼠精神分裂癥后可引起海馬齒狀回神經(jīng)細(xì)胞增殖的降低;(2)小鼠精神分裂癥后,在mRNA水平上,海馬GABAA的表達(dá)降低,NR1的表達(dá)升高。
[Abstract]:AIM: To investigate the co-existence pattern of NMDA receptor NR1 subunit, GABAA receptor and granulocyte layer of hippocampal dentate gyrus in schizophrenic (SP) mice, and to elucidate the expression of NR1 and GABAA receptors in the hippocampus of SP mice. METHODS: Mice in experimental group were injected with intraperitoneal injection of MK-801 (0.6 mg/kg daily), while those in control group were injected with the same amount of normal saline BrdU labeling was performed on the two groups of animals and the following treatment was performed: (1) Immunofluorescence staining was used to observe the expression of NR1 and GABAA in the DG region of the hippocampus and the coexistence pattern with BrdU labeled neogranular cells; (2) RT-PCR was used to detect the expression of GABAA and NR1 mRNA in the hippocampus. Compared with the control group, the proliferation rate of hippocampal neurons decreased by 23.1% (P 0.05), and there was no significant difference between GABAA and NR1 (P 0.05); (2) The expression of GABAA mRNA in the hippocampus of the experimental group was significantly lower than that of the control group (P 0.05), but the expression of NR1 mRNA was significantly higher than that of the control group (P 0.05). (2) After schizophrenia, the expression of GABAA in hippocampus decreased and the expression of NR1 increased.
