糖調(diào)節(jié)受損大鼠認(rèn)知障礙及腦組織NF-κBp65、TNF-α的表達

【摘要】:目的觀察糖調(diào)節(jié)受損大鼠腦組織NF-κBp65、TNF-α及NF-κB mRNA、TNF-αmRNA表達,探討糖耐量受損大鼠的炎癥機制及其對認(rèn)知功能障礙的影響。方法將Wistar大鼠隨機分為對照組和實驗組,高脂、高糖飼養(yǎng)法建立糖耐量受損大鼠模型;采用免疫組織化學(xué)和原位雜交的方法分別從蛋白水平和轉(zhuǎn)錄水平檢測糖調(diào)節(jié)受損組與糖耐量正常組大鼠腦組織NF-κBp65、NF-κB mRNA和TNF-α、TNF-αmRNA的表達;Morris水迷宮法評定兩組大鼠認(rèn)知功能。結(jié)果與NGT組比較,IGR組第10周NF-κBp65、NF-κB mRNA高表達;第15周TNF-α、TNF-αmRNA高表達。兩組間學(xué)習(xí)記憶能力在第5周無差異(P0.05);在第10周IGR組優(yōu)于NGT組;第15、第20周實驗組逐次下降(P0.05),低于同時點NGT組(P0.05)。Pearson相關(guān)分析顯示NF-κBp65、NF-κB mRNA和TNF-α、TNF-αmRNA高表達與大鼠認(rèn)知障礙相關(guān)。結(jié)論糖調(diào)節(jié)受損與大鼠認(rèn)知障礙相關(guān),NF-κB調(diào)控的炎癥反應(yīng)可能是其重要機制。
[Abstract]:Objective to investigate the expression of NF- 魏 B p65 TNF- 偽 and NF- 魏 B mRNA-tNF- 偽 mRNA in rats with impaired glucose tolerance, and to explore the inflammatory mechanism and its effect on cognitive impairment in rats with impaired glucose tolerance. Methods Wistar rats were randomly divided into two groups: control group and experimental group. The expression of NF- 魏 B p65 and TNF- 偽 TNF- 魏 B mRNA and TNF- 偽 TNF- 偽 mRNA in the brain of impaired glucose regulated group and normal glucose tolerance group were detected by immunohistochemistry and in situ hybridization respectively. The cognitive function of the two groups was evaluated by Morris water maze. Results compared with NGT group, the expression of NF- 魏 B p65 and TNF- 偽 mRNA in IGR group was higher than that in NGT group at 10 weeks and 15 weeks, respectively. There was no difference in learning and memory ability between the two groups at the 5th week (P0.05); IGR group was superior to NGT group at the 10th week; at the 15th and 20th week, the experimental group decreased one after another (P0.05), which was lower than NGT group (P0.05) .Pearson correlation analysis showed that the high expression of NF- 魏 B p65 and TNF- 偽 TNF- 偽 mRNA was associated with cognitive impairment in rats. Conclusion impaired glucose regulation and cognitive impairment may play an important role in the regulation of NF- 魏 B inflammation in rats.
