【摘要】：阿爾茨海默病(AD)是一種最常見的記憶和認知衰退的神經退行性疾病,影響了全世界數以百萬計的人,而目前對于AD的病因仍不明確且臨床上仍缺乏有效的治療手段。線粒體是細胞的能量工廠,線粒體功能障礙在衰老進程中起到的作用已經被越來越多的研究證實,這些功能異常包括氧化應激誘導產生的呼吸鏈功能異常、線粒體動力學缺陷、線粒體生物發(fā)生受損、mtDNA的突變等,其在AD的病理過程中扮演重要的角色,可作為神經保護作用的藥物治療靶點,即通過保護和提高線粒體功能來達到延緩或改善患者的臨床癥狀。本文主要闡釋了線粒體功能障礙在AD的主要表現,并介紹了一系列具有相當前景的線粒體靶點治療的藥物和技術方法,為阿爾茲海默病的治療提供了新的思路。
[Abstract]:Alzheimer's disease (AD) is one of the most common neurodegenerative diseases of memory and cognitive decline, affecting millions of people all over the world. Mitochondria are the energy factory of cells. The role of mitochondrial dysfunction in aging process has been confirmed by more and more studies. These abnormalities include respiratory chain dysfunction induced by oxidative stress and mitochondrial kinetic defects. Mitochondrial mutagenesis plays an important role in the pathological process of AD and can be used as a target of drug therapy for neuroprotective effect. That is by protecting and improving mitochondrial function to delay or improve the clinical symptoms of patients. This article mainly explains the main manifestation of mitochondrial dysfunction in AD, and introduces a series of promising drugs and technical methods of mitochondrial target therapy, which provides a new idea for the treatment of Alzheimer's disease.
