首發(fā)抑郁癥患者Notch通路及相關(guān)miRNAs的表達(dá)差異研究

【摘要】:目的:在人群水平探討Notch通路相關(guān)的m RNAs、mi RNAs與抑郁癥發(fā)生的關(guān)系。方法:采用病例對照研究設(shè)計(jì),嚴(yán)格按照入排標(biāo)準(zhǔn):1.實(shí)驗(yàn)一收集18名首發(fā)抑郁癥患者和18名正常人群的外周血,提取RNA,通過實(shí)時(shí)熒光定量PCR(Quantitative real-time PCR,q RT-PCR)檢測Notch信號(hào)通路中Notch1、RBP-jk、Numb、Hes1的m RNA表達(dá)水平,分析比較抑郁患者與正常人群外周血表達(dá)的差異、及其與認(rèn)知功能等指標(biāo)的相關(guān)性;2.實(shí)驗(yàn)二新收集32名首發(fā)抑郁癥患者和32名正常人群,采集外周血提取RNA,通過q RT-PCR檢測與Notch信號(hào)通路相關(guān)的mi RNAs(mi R-34b-5p、mi R-34c-5p、mi R-107、mi R-381、mi R-369-3p)以及m RNAs(Notch1、Hes1)的表達(dá)水平,分析比較抑郁癥患者與正常人群外周血表達(dá)的差異,及其與臨床癥狀、電生理、認(rèn)知功能等指標(biāo)的相關(guān)性。結(jié)果:實(shí)驗(yàn)部分一:1.抑郁組Notch1(Z=-3.354,P=0.001)表達(dá)低于對照組,RBP-jk(Z=-2.689,P=0.007)、Numb(Z=-3.101,P=0.002)表達(dá)高于對照組,未發(fā)現(xiàn)Hesl基因表達(dá)有差異(P0.05);2.抑郁癥患不同亞型分組后,Notch1、RBP-jk、Numb、Hes1表達(dá)無差異;未發(fā)現(xiàn)Notch相關(guān)因子與認(rèn)知功能存在相關(guān)性。實(shí)驗(yàn)部分二:1.抑郁組mi R-34b-5p(Z=-2.457,P=0.014)、mi R-34c-5p(Z=-2.001,P=0.045)表達(dá)高于正常組,抑郁組Notch1(Z=-2.021,P=0.043)的表達(dá)低于對照組;2.Notch1與mi R-34b-5p(r=-0.370,P=0.040)、mi R-34c-5p(r=-0.377,P=0.036)呈負(fù)相關(guān);在mi RNAs之間,mi R-34b-5p與mi R-34c-5p(r=0.881,P0.001)呈高度正相關(guān),mi R-34b-5p與mi R-107(r=0.794,P0.001)、mi R-381(r=0.877,P0.001)呈中、高度正相關(guān),mi R-34c-5p與mi R-107(r=0.943,P0.001)、mi R-381(r=0.892,P0.001)呈高度正相關(guān),mi R-107與mi R-381(r=0.835,P0.001)呈高度正相關(guān)。3.不同抑郁癥亞型分組后,有自殺意念的抑郁癥患者mi R-34b-5p(Z=-1.992,P=0.046)、mi R-369-3p(Z=-3.131,P=0.002)的表達(dá)都低于無自殺意念的患者。4.mi R-34b-5p(r=0.680,P0.001)、mi R-34c-5p(r=0.718,P0.001)、mi R-107(r=0.738,P0.001)、mi R-381(r=0.713,P0.001)與HAMD17評估得分呈正相關(guān);mi R-34b-5p(r=0.481,P=0.015)、mi R-34c-5p(r=0.506,P=0.010)、mi R-107(r=0.503,P=0.010)、mi R-381(r=0.485,P=0.014)與N1潛伏期呈正相關(guān),mi R-34b-5p(r=0.429,P=0.032)、mi R-34c-5p(r=0.479,P=0.016)、mi R-107(r=0.464,P=0.019)、mi R-381(r=0.566,P=0.003)與P2潛伏期呈正相關(guān)。結(jié)論:1.抑郁癥患者存在外周血Notch通路相關(guān)m RNAs表達(dá)異常。2.抑郁癥患者存在外周血Notch通路相關(guān)mi RNAs表達(dá)異常,且與抑郁癥臨床癥狀、認(rèn)知功能密切相關(guān)。3.mi R-34b-5p、mi R-34c-5p可能靶向調(diào)控Notch參與抑郁癥的發(fā)生。
[Abstract]:Objective: to investigate the relationship between Notch pathway related mRN Asmi RNAs and depression. Methods: a case-control study was designed, strictly according to the entry standard of 1: 1. In experiment 1, the peripheral blood of 18 patients with first-episode depression and 18 normal controls were collected, and the expression of m RNA in the Notch signaling pathway was detected by real-time fluorescence quantitative PCR(Quantitative real-time PCRQ RT-PCR1. To analyze and compare the difference of peripheral blood expression between depressive patients and normal subjects, and its correlation with cognitive function. In experiment 2, 32 first-episode depression patients and 32 normal controls were collected and the expression levels of mi RNAs(mi R-34b-5pttch1Hes1) and mRNAs-Notch1Hes1) related to Notch signaling pathway were detected by Q RT-PCR. To analyze and compare the difference of peripheral blood expression between depression patients and normal people, and its correlation with clinical symptoms, electrophysiology, cognitive function and so on. Results: the experiment part is 1: 1. The expression of Hesl gene in depression group was lower than that in control group. There was no significant difference in the expression of Numb-Hes1 in patients with different subtypes of depression, and no correlation between Notch related factors and cognitive function was found.
