Caspase-3抑制劑干預(yù)甲基苯丙胺慢性依賴大鼠腦神經(jīng)細(xì)胞凋亡與BCL-2表達(dá)的改變

【摘要】:目的觀察z-DEVD-fmk對甲基苯丙胺(MAP)依賴大鼠行為變化的影響;腦組織細(xì)胞Caspase-3蛋白酶活性的變化;腦組織相關(guān)區(qū)域神經(jīng)細(xì)胞凋亡及對BCL-2蛋白表達(dá)的影響。 方法建立甲基苯丙胺依賴大鼠實(shí)驗(yàn)動物模型,同時側(cè)腦室注射z-DEVD-fmk進(jìn)行干預(yù),采用酶標(biāo)儀法檢測Caspase3酶活性;TUNEL法檢測大鼠腦神經(jīng)細(xì)胞凋亡;免疫組化技術(shù)檢測BCL-2蛋白表達(dá)的影響。 結(jié)果 1.MAP慢性依賴大鼠100%出現(xiàn)異常行為表現(xiàn),主要有異常不自主點(diǎn)頭、豎毛自殘、原地轉(zhuǎn)圈;易激動等癥狀均有發(fā)生,給予z-DEVD-fmk干預(yù)后上述癥狀發(fā)生較少。 2. Caspase3酶活性:z-DEVD-fmk干預(yù)后與MAP慢性依賴組相比較Caspase-3酶活性降低,有顯著性差異(P<0.05)。 3.大鼠腦組織細(xì)胞凋亡:z-DEVD-fmk干預(yù)下的MAP慢性依賴組大鼠腦組織細(xì)胞凋亡明顯減少,與MAP慢性依賴組相比有顯著性差異(P<0.01)。 4. BCL-2蛋白表達(dá):免疫組織化學(xué)染色顯示z-DEVD-fmk干預(yù)下,,MAP慢性依賴組大鼠腦組織細(xì)胞BCL-2陽性細(xì)胞數(shù)增加,與MAP慢性依賴組相比較有顯著性差異(P<0.01)。 結(jié)論 1. z-DEVD-fmk干預(yù)后Caspase-3酶活性較MAP組降低; 2. z-DEVD-fmk干預(yù)后減少M(fèi)AP慢性依賴大鼠腦組織細(xì)胞凋亡,上調(diào)BCL-2蛋白的表達(dá)。
[Abstract]:Objective to observe the effect of z-DEVD-fmk on the behavioral changes of rats with methamphetamine mitogen-dependent activity of Caspase-3 protease in brain tissue and the effect of neuronal apoptosis on the expression of BCL-2 protein in brain tissue related regions. Methods the experimental animal model of methamphetamine dependent rats was established, and z-DEVD-fmk was injected into the lateral ventricle to intervene. The activity of Caspase3 was detected by enzyme labeling method and Tunel method was used to detect the apoptosis of rat brain nerve cells, and the effect of BCL-2 protein expression was detected by immunohistochemical technique. Result Abnormal behavior was found in 100% of 1.MAP chronic dependent rats, including abnormal involuntary nodding, self-mutilation of hair, rotation in situ, agitation and other symptoms, and the occurrence of these symptoms was less after the intervention of z-DEVD-fmk. 2. The activity of Caspase3 enzyme: z-DEVD-fmk was significantly lower than that of chronic dependent MAP group (P < 0.05). 3. Apoptosis of brain tissue cells in MAP chronic dependent group was significantly decreased after intervention of z-DEVD-fmk (P < 0.01), which was significantly lower than that in MAP chronic dependence group (P < 0.01). 4. Expression of BCL-2 protein: immunohistochemical staining showed that the number of BCL-2 positive cells in brain tissue of rats with chronic dependence of map was increased after z-DEVD-fmk intervention, and there was a significant difference compared with that of chronic dependence group of MAP (P < 0.01). Conclusion 1. The activity of Caspase-3 in z-DEVD-fmk group was lower than that in MAP group. 2. After z-DEVD-fmk intervention, the apoptosis of brain cells and the expression of BCL-2 protein were decreased in chronic dependent MAP rats.
