【摘要】：目的：探討炎癥因子IL-1α基因(rs1800587)、IL-1β基因(rs1143627)、IL-18基因(rs187238)和IL-33基因(rs11792633)單核苷酸多態(tài)性(SNP)位點與湖南漢族人群遲發(fā)型阿爾茨海默病(LOAD)的關(guān)系。 方法：運用病例-對照研究,選取LOAD患者201例為AD組,另選擇年齡及性別相匹配的健康人群257例為對照組。利用聚合酶鏈反應(yīng)(PCR)和基質(zhì)輔助激光解吸電離飛行時間質(zhì)譜技術(shù)(MALDI-TOF-MS)對炎癥因子IL-lα、IL-1β、IL-18、IL-33共四個SNPs位點(rs1800587、rs1143627、rs187238和rs11792633)進行基因分型檢測,比較不同基因型、等位基因頻率在兩組間的差別,并分析四個SNPs位點與LOAD的相關(guān)性。 結(jié)果： 1.IL-1α基因rs1800587各基因型頻率在兩組間差異無統(tǒng)計學(xué)意義(P0.05),CC型和(CT+TT)型及C、T等位基因頻率在兩組間差異有統(tǒng)計學(xué)意義(P0.05),攜帶T等位基因者具有較高的LOAD發(fā)病風(fēng)險(OR=1.592,95%CI=1.037-2.443)。 2.IL-1β基因rs1143627及IL-18基因rs187238各基因型及等位基因頻率在兩組間差異無統(tǒng)計學(xué)意義(P0.05)。 3.IL-33基因rs11792633各基因型及等位基因頻率在兩組間差異有統(tǒng)計學(xué)意義(P0.05),攜帶T等位基因者具有較低的LOAD發(fā)病風(fēng)險(OR=0.648,95%CI=0.498-0.843)。 4.多因素非條件Logistic回歸分析示文化程度、喪偶、總膽固醇、IL-1α基因rs1800587及IL-33基因rs11792633與LOAD顯著相關(guān)。 結(jié)論： 1.IL-1α基因rs1800587位點多態(tài)性與湖南漢族人群LOAD易感性相關(guān)聯(lián),T等位基因可能是湖南漢族人群LOAD的危險因素。 2.IL-1β基因rs1143627位點多態(tài)性和IL-18基因rs187238位點多態(tài)性與湖南漢族人群LOAD易感性無關(guān)聯(lián)。 3.IL-33基因rs11792633位點多態(tài)性與湖南漢族人群LOAD易感性相關(guān)聯(lián),T等位基因可能是湖南漢族人群LOAD的保護因素。
[Abstract]:Objective: to investigate the relationship between the single nucleotide polymorphisms (SNPs) of the inflammatory factor IL-1 偽 gene rs1800587 and IL-33 gene rs11792633) and the late onset Alzheimer's disease (Lod) in Hunan Han population. Methods: a case-control study was conducted in which 201 patients with LOAD were selected as AD group and 257 healthy subjects matched with age and sex as control group. Polymerase chain reaction (PCR) and matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) were used to detect four SNPs loci (rs1800587rs1143627rs187238 and rs11792633) of inflammatory factor IL-l 偽, IL-1 尾 and IL-18IL-33. The correlation between four SNPs loci and LOAD was analyzed. Results: There was no significant difference in the frequency of rs1800587 genotypes of 1.IL-1 偽 gene between the two groups. There was no significant difference between the two groups in the frequency of rs1800587 genotypes (P0.05C and TTCT) and the frequencies of CT-alleles were significantly different between the two groups. The patients with T alleles had a higher risk of LOAD. There was no significant difference in the frequency of 2.IL-1 尾 rs1143627 and IL-18 rs187238 genotypes and alleles between the two groups (P 0.05). The frequencies of rs11792633 genotypes and alleles of 3.IL-33 gene were significantly different between the two groups (P 0.05). Those with T allele had a lower risk of LOAD. 4. Multivariate conditional Logistic regression analysis showed that education level, widowed degree, total cholesterol IL-1 偽 gene rs1800587 and IL-33 gene rs11792633 were significantly correlated with LOAD. Conclusion: The rs1800587 polymorphism of 1.IL-1 偽 gene is associated with the susceptibility to LOAD in Hunan Han population. The T allele may be the risk factor of LOAD in Hunan Han population. The polymorphism of 2.IL-1 尾 gene rs1143627 locus and IL-18 gene rs187238 locus were not associated with LOAD susceptibility in Hunan Han population. The polymorphism of 3.IL-33 gene rs11792633 locus is associated with the susceptibility to LOAD in Hunan Han population. The T allele may be the protective factor of LOAD in Hunan Han population.
