急性創(chuàng)傷應(yīng)激對小鼠行為變化的影響

【摘要】:目的研究急性創(chuàng)傷應(yīng)激對小鼠自發(fā)行為、焦慮、學(xué)習(xí)記憶的影響。方法將16只小鼠分為實驗組(n=8)和對照組(n=8),實驗組給予足底電擊,對照組不給予任何處理;分別在處理后0 h、12 h、7 d進(jìn)行曠場實驗以及高架十字迷宮實驗,以評價小鼠行為學(xué)改變;用水迷宮實驗以評價小鼠學(xué)習(xí)記憶改變。結(jié)果 (1)兩組小鼠的站立次數(shù)和中央?yún)^(qū)域活動時間、路程在應(yīng)激后0 h和12 h差異均有統(tǒng)計學(xué)意義(P0.01),實驗組少于對照組;應(yīng)激后7 d兩組僅中央活動時間差異有統(tǒng)計學(xué)意義(P0.05);(2)實驗組小鼠進(jìn)入開臂次數(shù)及停留時間在應(yīng)激后立即、12 h較對照組均顯著減少(P0.05);(3)實驗組小鼠第2、3天的逃避潛伏期較對照組顯著延長(P0.05),穿越平臺次數(shù)及目標(biāo)象限游程比例較對照組均顯著增多(P0.05)。結(jié)論足底電擊可引起小鼠自發(fā)行為減少,導(dǎo)致探索樣行為減弱,焦慮增加,學(xué)習(xí)能力降低,但在一定程度上增強(qiáng)小鼠的空間記憶能力。
[Abstract]:Objective to study the effects of acute traumatic stress on spontaneous behavior, anxiety, learning and memory in mice.Methods Sixteen mice were divided into two groups: the experimental group and the control group. The experimental group was given electric shock on the plantar, the control group was given no treatment, the open field test and the elevated cross maze test were carried out at 12 hours and 7 days after treatment to evaluate the behavior of the mice.Water maze test was used to evaluate the changes of learning and memory in mice.Results (1) the number of times of standing and the time of central area activity in the two groups were significantly different between the two groups at 0 h and 12 h after stress, and the experimental group was less than the control group.7 days after stress, there was a significant difference in the time of central activity between the two groups (P 0.05) the number of times of entering the arm and the staying time of the mice in the experimental group immediately after stress decreased significantly compared with the control group for 12 h) the mice in the experimental group escaped from latent latency on the 2nd day of the third day.Compared with the control group, the time of crossing the platform and the proportion of the target quadrant run were significantly longer than that of the control group (P 0.05).Conclusion the plantar electric shock can reduce the spontaneous behavior, weaken the exploratory behavior, increase anxiety and decrease the learning ability, but to some extent enhance the spatial memory ability of mice.
