丁苯酞對血管性癡呆大鼠海馬CA1區(qū)腦源性神經(jīng)營養(yǎng)因子表達(dá)的影響

【摘要】:目的:觀察丁苯酞(NBP)對血管性癡呆(VD)大鼠海馬CA1區(qū)腦源性神經(jīng)營養(yǎng)因子(BDNF)表達(dá)的影響,探討丁苯酞對VD的保護(hù)作用。方法:將80只健康Wistar大鼠按隨機(jī)區(qū)組法分為假手術(shù)組、NBP對照組(假手術(shù)+NBP注射劑)、VD組(VD模型)和NBP處理組(VD模型+NBP注射劑),每組20只,每組又分為4個亞組,即術(shù)后1、2、4和8周組,每個亞組5只。永久性雙側(cè)頸總動脈結(jié)扎法制備VD大鼠模型。NBP對照組和NBP處理組大鼠腹腔注射NBP注射劑5mg·kg~(-1)·d~(-1),連續(xù)給藥7d。假手術(shù)組和VD組大鼠每次腹腔注射4mL生理鹽水,連續(xù)注射7d。各組大鼠在術(shù)后各時間點(diǎn)(1、2、4和8周)留取海馬組織,應(yīng)用實(shí)時定量PCR和免疫組織化學(xué)法檢測各組大鼠海馬CA1區(qū)BDNF表達(dá)水平。結(jié)果:定時定量PCR法,術(shù)后2、4和8周,VD組大鼠海馬BDNF mRNA表達(dá)水平明顯高于假手術(shù)組(P0.05),術(shù)后4和8周,NBP處理組大鼠海馬BDNF mRNA表達(dá)水平明顯高于VD組(P0.05);免疫組織化學(xué)法,VD組大鼠4周時BDNF表達(dá)水平明顯高于假手術(shù)組(P0.05),術(shù)后8周時NBP組大鼠CA1區(qū)BDNF蛋白表達(dá)水平明顯高于VD組(P0.05)。結(jié)論:VD模型大鼠海馬CA1區(qū)神經(jīng)元在遭受缺血損傷后,BDNF反應(yīng)性表達(dá)增加,而NBP則可明顯提高VD大鼠海馬CA1區(qū)BDNF的表達(dá),發(fā)揮神經(jīng)保護(hù)作用。
[Abstract]:Aim: to observe the effect of butyphthalide (NBP) on the expression of brain-derived neurotrophic factor (BDNF) in CA1 area of vascular dementia rats. To investigate the protective effect of butyphthalide on VD. Methods: 80 healthy Wistar rats were randomly divided into two groups: sham operation group (n = 20) and NBP treated group (n = 20). Each group was subdivided into 4 subgroups, namely, 1, 2, 4 and 8 weeks after operation. Five rats in each subgroup were used to establish VD rat model by permanent bilateral common carotid artery ligation. Rats in the control group and the NBP treatment group were injected intraperitoneally with NBP injection 5mg KGG -1) for 7 days. Rats in sham operation group and VD group were intraperitoneally injected with 4mL saline. After 7 days of continuous injection, the hippocampal tissue was taken from the rats of each group at each time point. The expression of BDNF in the hippocampal CA1 was detected by real-time quantitative PCR and immunohistochemical method. Results: the expression of BDNF in the hippocampus of the rats in each group was measured by quantitative PCR method. The expression of BDNF mRNA in hippocampus of VD group was significantly higher than that of sham operation group at 2 and 8 weeks postoperatively, and the expression of BDNF mRNA in hippocampus of VD group was significantly higher than that of VD group at 4 and 8 weeks after operation, and the expression of BDNF in VD group was significantly higher than that in VD group at 4 weeks and 8 weeks after operation, and the expression of BDNF mRNA in VD group was significantly higher than that in VD group at 4 weeks after operation. The expression of BDNF protein in CA1 was significantly higher in NBP group than that in VD group at 8 weeks after operation. Conclusion the reactive expression of BDNF in hippocampal CA1 neurons of rats with NBP was increased after ischemia injury. NBP could significantly increase the expression of BDNF in hippocampus CA1 of VD rats and play a neuroprotective role.
